基于核磁共振技术的模式识别方法在肾气虚哮喘大鼠血清代谢组学中的应用 被引量:9

摘要 目的:分析肾气虚哮喘大鼠血清代谢图谱的变化,探讨代谢组学在肾气虚哮喘研究中的应用。方法:将56只大鼠随机分成正常对照组、肾气虚哮喘模型组、西药组、中药组,进行哮喘、肾气虚哮喘造模及中西药治疗,观察治疗前后大鼠症状、体征变化。造模成功后经超导核磁共振波谱仪Bruker 600MHz采集大鼠血清代谢氢谱图,结合主成分分析法(PCA)、偏最小二乘法判别分析法(PLS-DA)和正交偏最小二乘法判别分析法(OPLSDA)进行模式识别分析。结果:各组大鼠血清代谢存在显著差异。结论:可通过一维氢谱核磁共振(1H-NMR)寻找潜在的肾气虚哮喘生物标志物。 Objective:To analysis the serum metabolic patterns changes of kidney-qi deficiency asthma rats,and to discuss the application of metabonomics in the kidney-qi deficiency asthma research.Methods:Fifty-six rats were randomly divided into a normal control group,kidney qi deficiency asthma group,western medicine group and Chinese medicine group.Multiple factors were used to establish asthma model and kidney-qi deficiency asthma model,which were treated by traditional Chinese medicine and western medicine.At the same time,we observed the changes of symptoms and signs in rats before and after treatment.After modeling,serum metabolites Hydrogen spectrum were obtained by Bruker 600 mHz NMR and was analyzed for pattern recognition through principal component analysis(PCA),partial least squares discriminant analysis(PLS-DA) and orthognalpartial least squares discriminant analysis(OPLS-DA) method.Results:All groups of rats serum metabolic existed significant differences.Conclusion:Potential biomarkers of kidney-qi deficiency asthma can be found through ~1H-NMR.
