摘要
测定毛皮软化液中的酶活力是实现毛皮软化液循环使用的基础。结合毛皮生产软化废水的组成及特点,研究了氯化钠、氨基酸对蛋白酶活力的影响。根据现有福林法测定蛋白酶活力的原理和特点,优化了软化废液透析预处理及酶活力测定条件,提出了"透析预处理-福林法测定软化废液酶活力"的方法。结果表明:软化废液中的食盐和酪氨酸对测定结果干扰较大,需要予以排除;透析预处理-福林法测定软化废液酶活力的加标回收率在100.0%~110.0%之间,能够基本满足工业化测定的要求。研究成果对毛皮企业在毛皮软化液循环使用中酶活力定量具有实际指导意义。
Determination of enzyme activity is the basis of realizing the fur bating wastewater recycling. The effect of sodium chloride, amino acid on protease activity was studied based on the components and characteristics of fur bating wastewater. The method of "dialysis pretreatment - forint phenol testing the enzyme activity of bating wastewater" was put forward by optimizing the testing conditions of dialysis pretrcatment and enzyme activity based on the testing theory and characteristics of protease activity of current forint phenol method. Results show that the sodium chloride and tyrosine in bating wastewater can severely impact the testing results, needing to be excluded. Recoveries of the dialysis pretreatment - forint phenol method range from 100.0% to 110.0%, which can basically meet the requirements of industrial determination. The research achievements have practical guiding significance in enzyme activity ration of recycling fur bating wastewater.
