摘要
BRAF是RAF基因家族成员,主要存在于神经组织和睾丸组织中,通过Ras/Raf/Mek/Erk信号联级通路调节细胞生长、增殖和凋亡。BRAF基因突变在多种肿瘤如大肠癌、甲状腺乳头状癌及黑色素瘤等肿瘤的发生、发展中起重要作用。该基因突变在胶质瘤中有明显表达,不同突变方式对胶质瘤的影响不同,BRAF V600E抑制剂为胶质瘤的治疗提供了新靶点。本文就BRAF基因的突变方式、突变机制以及BRAF V600E抑制剂在胶质瘤治疗中的作用等研究进展作一综述。
BRAF is a member of the RAF gene family,mainly existing in nerve system and testicular tissue.It regulates cell growth,proliferation and apoptosis through Ras/Raf/Mek/Erk signaling pathway.It has been proved that BRAF mutation plays an important role in the occurrence and development of various tumors,as colorectal cancer,thyroid papillary carcinoma and melanoma.The latest study has found that BRAF gene mutation is obviously expressed in glioma,and different ways of mutations have different influence on glioma.The BRAF V600 Einhibitor also provides a new target for the treatment of glioma.This paper reviews the style and mechanism of BRAF mutation,as well as the role of BRAF V600 Einhibitor in the treatment of gliomas.
