清脑灵诱导大鼠骨髓间充质干细胞分化成神经元样细胞

摘要 目的:观察清脑灵煎剂是否能诱导大鼠骨髓间充质干细胞(MSCs)分化成神经元样细胞。方法:将21只四周SD大鼠依照随机性原则分成三组,即空白对照组、β-巯基乙醇组以及清脑灵组,各组均为7只。通过贴壁法分离获得大鼠骨髓间充质干细胞,体外扩增培养,用流式细胞仪检测细胞表面抗原的表达。清脑灵组用浓度为1mg/m L的清脑灵煎剂诱导MSCs,而β-巯基乙醇组则使用β-巯基乙醇诱导MSCs,空白对照组不进行任何处理,分别于5h、12h、1d、3d、7d在倒置显微镜下观察细胞形态变化,用免疫细胞化学方法观察被诱导细胞巢蛋白(Nestin)、神经元特异性烯醇化酶(NSE)、神经胶质纤维酸性蛋白(GFAP)的表达。结果:流式细胞仪检测显示MSCs不表达CD11b、CD45,诱导后细胞的免疫细胞化学示Nestin阳性细胞、NSE阳性细胞和GFAP阴性细胞。结论:清脑灵能诱导MSCs分化成神经元样细胞。 Objective: To observe inducing differentiation of rats MSCs into neuron-like cells with Qingnaoling. Methods: 21 rats were divided into the control group, the β-mercaptoethanol group and the Qingnaoling group, 7 rats in each. MSCs were isolated by adherent method and cultured in vitro. The expression of cell surface antigens was detected by flow cytometry. The Qingnaoling group was given Qingnaoling (1 mg/mL); the fl-mercaptoethanol group was given β-mercaptoethanol. The nestin, NSE, GFAP of the induced cells were observed by immunocytochemistry. Results: The expression of CD11b and CD45 in MSCs was not detected by flow cytometry. In inducing differentiation, the nestin-positive cells, NSE-positive cells, GFAP-negative cells of the induced were detected. Conclusion: Qingnaoling can induce differentiate of MSCs into neuron-like cells.
