摘要
目的研究家兔脑干机械性损伤后S100β含量随时间变化规律。方法健康家兔24只,经自制打击装置打击脑干部位,分别于损伤后0、2、4、8、16、24、48、72h解剖取脑干,甲醛固定,免疫组织化学和图像分析技术检测。结果损伤后Oh家兔脑干中可见S100β阳性染色均匀分布在星形胶质细胞胞体及突起中。脑干机械性损伤后2h,星形胶质细胞结构破坏,大量的S100β从中漏出,突起消失;之后随着时间的延长S100β逐步升高,24h达到高峰,之后下降,但仍高于Oh(LSD检验,P〈0.05)。结论S100B主要存在于神经胶质细胞,是中枢神经损伤的敏感生化指标,根据S100β的含量变化可作检测脑早期缺血缺氧性损伤和预后的黄金指标;同时可以应用于法医学中枢神经系统损伤时间的推断,为法医学鉴定提供一定的实验依据。
Objective To study the regulation of time-dependent expression of S100β following brain stem mechanical injury in rabbits. Methods The brain stem of 24 health rabbits injuried by self-made devices are dissected after injury 0,2,4,8,16,24,48, 72h. then formalin fixation and detected by immunohistochemistry and auto-image analysis. Results positive stain of S100β uniformly distributing in cell body and apophysis of astroglia cells in brain stem of rabbits after injury Oh. After mechanical injury of brain stem 2h, the destroyed astroglia cells release mass S100β with apophysis dispearing, then with time pass by the concetration of S100β increasing,at 24h after injury the concetration reachs the peak. Then the eoncetration decreased but is higher than that at Oh (LSD analysis,P〈0.05). Conclusion S100β mainly lies in neuroglial cell,is sensitive biochemical index and is golded index of early hypoxic-ischemic brain damage and prognosis. Otherwise S100β also apply for concluding injury age of forensic injury of central nervous system, so offer experiment basis for the forensic identification.
