薄层色谱-生物自显影联用检测残留抗生素的研究 被引量:8

Detection of Residual Antibiotics in Agricultural and Sideline Products by HPTLC Coupled with Bioluminescence

摘要 建立利用薄层色谱-生物自显影联用原位技术检测农副产品中的抗生素残留方法。采用饱和的EDTA-2Na溶液活化的硅胶薄层层析板,展开溶剂系统为乙酸乙酯-氯仿-甲醇-氨水(40∶40∶20∶5),于紫外灯下记录薄层色谱图,以发光细菌费氏弧菌(V.fi scheri)为检定菌,薄层板表面均匀涂布处于最强发光周期的费氏弧菌(V.fi scheri)发酵液,放入生物发光检测仪中,拍照记录。结果表明,100份样品中的72份被检出有残留抗菌物质存在,并对残留抗生素进行初步推定。该方法操作简便、快捷、结果准确,可进一步研究用于抗生素残留定性与定量检测。 Establish a method to detect the residual antibiotics in agricultural and sideline products by HPTLC coupled with bioluminescence. The silica thin layer was treated by saturation EDTA-2Na solution, samples was separated on silica thin layer plate with mixture solvent of EtOAc/CHCl3/CH3OH/ammonia water (40 : 40 : 20 : 5), chromatogram was recorded under UV254 nm and UV365nm, The luminous V. fischeri bacterial liquid was coated uniformly on the developed thin layer plate, V. fischeri was used as test bacterium. The TLC plate was put into Bioluminizer for detection, and then recorded by photographing. The result showed that 72 samples were detected with antibiotic materials among a total of 100 samples, residual antibiotics were determined preliminarily. This method was rapid, accurate and reliable, it was worth a further study to be used as the identification and determination method of residual antibiotics.
