ADAR1对口腔鳞癌细胞生物学特征的影响

摘要 检测RNA编辑酶1(ADAR1)在口腔鳞癌细胞中的表达,探讨其对口腔鳞癌细胞生物学行为特征的影响及作用机制。方法:利用实时定量RT-PCR、Western Blot检测ADAR1在口腔鳞癌细胞中的表达;化学合成siRNA(si-ADAR1)转染细胞,观察其迁移和侵袭能力;检测ADAR1敲减后口腔鳞癌细胞中上皮间质转化指标(Vimentin、E-cadherin)、干性指标(Sox2、Oct4)以及onco-microRNAs(miR-21-3p、miR-18a-3p、miR-210-3p、miR-155-5p、miR-181a-3p、miR-19a-3p)表达水平。结果:ADAR1在口腔鳞癌细胞中高表达;ADAR1敲减后的口腔鳞癌细胞迁移和侵袭能力下降(P<0.05),上皮间质转化指标E-cadherin低表达、Vimentin高表达,干性指标(Sox2、Oct4)及口腔鳞癌相关onco-MicroRNAs低表达。结论:ADAR1与口腔鳞癌细胞的生物学行为密切相关,推测ADAR1可能通过促进口腔鳞癌相关onco-microRNAs成熟影响口腔鳞癌细胞的生物学行为。 Objective:To examine the expression of adenosine deaminases acting on RNA 1(ADAR1) in oral squamous cell carcinoma(OSCC) and its influence on biological behavior of OSCC.Methods:The expression of ADAR1 in OSCC cell lines was tested by RT-q PCR and Western Blot.The expression of ADAR1 gene in OSCC cell lines was knocked down by si ADAR1.Wound healing and Transwell test were used to detect migration and invasion of OSCC.RT-q PCR and Western Blot were used to detect epithelial-mesenchymal transition(EMT) markers Vimentin and E-cadherin,stemness markers Sox2 and Oct4,and OSCC-associated onco-microRNAs(miR-21-3p,miR-18a-3p,miR-210-3p,miR-155-5p,miR-181a-3p,miR-19a-3p).Results:Higher ADAR1 was expressed in OSCC compared to normal oral mucosa.After ADAR1 knocked down,not only the migration and invasion of OSCC were significantly inhibited(P〈0.05),but also EMT and stemness markers such as Vimentin,Sox2,and Oct4 decreased.Of note,OSCC-associated onco-microRNAs(miR-21-3p,miR-18a-3p,miR-210-3p,miR-155-5p,miR-181a-3p,miR-19a-3p) also decreased with ADAR1 gene knock down.Conclusions:ADAR1 is closely related with the biological behavior of OSCC.We speculate that ADAR1 may have impact on the biological behavior of OSCC by promoting mature microRNA processing.
