超声波辅助法提取万寿菊中叶黄素及其抗动脉粥样硬化研究 被引量:5

摘要 目的:优化叶黄素的提取工艺,并初步探讨了叶黄素对实验性高脂家兔的降脂及抗动脉粥样硬化(AS)作用。方法:采用正交设计法优化叶黄素提取工艺,高脂饲料喂养家兔建立高脂血症和动脉粥样硬化模型。使用叶黄素连续给药4周,采用试剂盒分别测定血清总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白(HDL-C)、低密度脂蛋白胆固醇(LDL-C)及门冬氨酸氨基转移酶(AST)、肌酸激酶(CK)、血浆内皮素(ET)、一氧化氮(NO)和环磷酸鸟苷(c GMP)含量,取主动脉,观察动脉斑块面积并进行病理学检查。结果:通过正交实验后得出最佳实验条件下叶黄素提取率最高可达90%以上;叶黄素高剂量组能降低家兔血清中TC、TG、LDL-C含量(P﹤0.05),升高血浆NO、c GMP水平(P﹤0.05),减少ET生成(P﹤0.05),降低主动脉粥样硬化病理变化。结论:采用超声提取可以提高叶黄素的提取效率,叶黄素通过调节血脂代谢、升高血浆NO、c GMP含量,减少ET生成等方式达到抗动脉粥样硬化的作用。 Objective To optimize the technology of extracting xanthophylls and initially study the action of extractive at preserving AS.Methods Orthogonal design was applied to get Determining,the hyperlipemia atherosclerosis models of rabbit were establishedby high fat diet. Gastric perfusion of Xanthophylls for four weeks. The content of TC、TG、LDL-C、HDL-C、AST、CK 、ET、NO、c GMP were determined. The extent of aortic atherosclerosis was observed and the pathological examination was done. Results Using this experiment conditions,the extracting ratio was more than 90%. Xanthophylls significantly decreased the level of TC,TG and LDL- C( P ﹤ 0.05),upregulated the level of plasma NO and c GMP( P ﹤ 0. 05),and cut down the pathological score of coronary atherosclerosis. Conclusion Using our experiment conditions to extract xanthophylls,Xanthophylls has antiatherosclerotic effects on atherogenesis in rabbits with hyperlipoidemia through regulating the blood lipid metabolism,increasing the levels of plasma NO and c GMP and decreasing the level of plasma ET.
