摘要
为明确天冬酰胺内肽酶(legumain)基因在斑马鱼发育中的表达特性,利用全胚胎原位杂交的方法检测legumain在斑马鱼胚胎发育过程中的表达分布情况,并采用双色原位杂交的方法研究legumain和泛髓系细胞、巨噬细胞、中性粒细胞的共定位情况。结果显示,legumain转录本为母系表达,在胚胎发育早期泛在性表达。在受精18h后在造血组织有特异的高丰度表达。双色原位杂交显示,legumain和泛髓系细胞、巨噬细胞、中性粒细胞有共定位表达。结果提示legumain在斑马鱼发育过程中有重要作用,在巨噬细胞中高表达并参与相关生理功能的完成。
To explore the spatio-temporal expression pattern of legumain in the zebrafish development, whole-mount in situ hybridization (WISH) was used to detect the expression pattern of legumain in the process of zebrafish embryonic development.Double in situ hybridization (D-WISH) was performed to detect the co-localization of legumain and pan-myeloid cells/macrophages/ neutrophiles.Legumain transcript was maternal expressed,and expressed ubiquitously in most tissues of early embryos.Then legumain transcript was specific highly expressed in hematopoietic tissue from 18 hour post fertilization (hpf).Legumain was found to co-localization with pan-myeloid cells/ macrophages/ neutrophiles using double in situ hy- bridization.The present study provided important clues that legumain plays an important role in embryonic development in zebrafish,and has high expression in macrophages and participates macrophage functions.
