新型鸭Ⅲ型腺病毒的分离鉴定及遗传进化分析 被引量:5

摘要 从广东某鸭场发病鸭体内分离到1株鸭Ⅲ型腺病毒(DA-1株),该病毒能在鸭成纤维细胞上繁殖,并呈现典型的细胞病变。通过高通量测序,获得了鸭Ⅲ型腺病毒(DA-1株)的全基因组序列。同源性分析表明,DA-1分离株hexon和DBP基因氨基酸序列与2014年法国分离的鸭Ⅲ型DA GR株的同源性最高,分别为86.2%和87.9%。遗传进化分析表明,DA-1株的hexon和DBP基因与GenBank上参考毒株相比,与鸭Ⅲ型DA GR株亲缘关系最近。鸭Ⅲ型DA是一种新出现的病毒,对养鸭业造成了一定的经济损失,研发高效疫苗和快速诊断方法对防控该病的流行具有重要的意义。 An adenovirus type Ⅲ virus named DA-1 was isolated from the ill ducks of a duck facto- ry in Guangdong, which could cause obvious cytopathic effect. High-throughput sequencing was used to obtain the complete gene of DA-1. The deduced amino acid sequences of hexon and DBP gene had the highest homology with the duck adenovirus typeⅢ GR isolated in France,and the homology was 86.2% and 87. 9%, respectively. Compared with other reference isolates in Gen- Bank,Phylogenetic analysis of the hexon and DBP gene of DA-1 isolate showed that the DA-1 isolate was most closest with GR isolate. The duck adenovirus typeⅢ is a new emerging virus which caused economic loss to duck farms, developing an effective vaccine and rapid diagnostic method are essential to control the epidemiology of the new virus.
