摘要
目的探究电针治疗缓解期偏头痛的可能作用机制及腧穴配伍的效应。方法将30只SD大鼠随机分为对照组、模型组、单穴组、配穴组、假穴组,每组6只。除对照组外其余各组大鼠颅顶钻孔植入电极手术后电刺激硬脑膜建立缓解期偏头痛模型。单穴组电针双侧风池穴,配穴组电针双侧风池穴加双侧阳陵泉穴,假穴组取大鼠腰部非穴位点,于造模后第2、4、6天实行清醒状态下电针干预,并测定大鼠足底和面部痛阈值,第7天检测大鼠脑三叉神经节、三叉神经脊束核尾核、中缝大核中5-羟色胺1F(5-HT1F)受体mRNA及蛋白表达。结果第2、4、6天模型组大鼠面部、足底痛阈值均明显低于同时间对照组(P<0.05);单穴组和配穴组面部、足底痛阈值均高于同时间模型组(P<0.05);单穴组面部、足底痛阈值低于同时间配穴组而高于同时间假穴组(P<0.05)。模型组大鼠脑三叉神经节、三叉神经脊束核尾核、中缝大核中5-HT1F受体mRNA及蛋白表达明显低于对照组(P<0.05);单穴组和配穴组各部位5-HT1F受体mRNA及蛋白表达明显高于模型组,并且配穴组高于单穴组(P<0.05)。结论电针治疗缓解期偏头痛机制可能与5-HT1F受体激活作用有关,且远近配穴疗效优于单穴治疗。
Objective To explore the possible mechanism of electroacupuncture in treating remission migraine and the effect of acupoints compatibility. Methods Thirty rats were randomized into a control group, a model group, a single point group, a compatibility points group, and a sham points group, with 6 rats in each group. Besides the control group, remission migraine models were established in rats in other groups by electrical stimulation of the dura mater after surgery of electrode operation for intracranial apical drilling. The single point group was given electroacupuncture at bilateral PointFengchi (GB20). The compatibility points group was given electroacupuncture at bilateral Points Fengchi (GB20) and Yanglingquan (GB34). The sham points group was given electroacupuncture at non acupoints at the waist. On the 2nd, 4th and 6th day after modeling, rats were given electroacupuncture under conscious state. Pain threshold value of pelma and face was examined. On the 7th day, the expressions of 5-hydroxytryptamine 1 F receptor (5-HT1F) mRNA and proteins in rats brain trigeminal ganglion, spinal trigeminal caudal nucleus and nucleus raphe magnus were detected. Results On the 2nd, 4th and 6th day, the pain thresh- old value of face and pelma of rats in the model group was lower than that in the control group at the same time ( P 〈 0. 05 ), that in the single point group and the compatibility points group were higher than that in the model group at the same time ( P 〈 0. 05), and that in the single point group was lower than that in the compatibility points group at the same time but higher than that in the sham points group at the same time ( P 〈 0.05 ). The expressions of 5-HT1F receptor mRNA and proteins in brain trigeminal ganglion, spinal trigeminal caudal nucleus and nucleus raphe magnus of rats in the model group were lower than those in the control group ( P 〈 0. 05 ) ; 5-HT1F receptor mRNA and pro- teins of all the sites in the single point group and the compatibility points group were higher than those in the model group, and those in the compatibility points group were higher than those in the single point group (P 〈 0. 05 ). Conclusion The mechanism of electroacupuncture activation. The effect of near-far point compatibility treating remission migraine might be related to 5-HT1 F receptor seems superior to single point treatment.
