摘要
目的建立高效液相色谱法测定大鼠血浆中的米卡芬净浓度,并考察其在大鼠体内的药动学。方法使用Diamonsil C_(18)色谱柱,流动相为乙腈:10mmol/L醋酸铵溶液(42.5:57.5,V/V),检测波长269nm,进行专属性、标准曲线、定量下限、精密度、回收率与稳定性考察。结果米卡芬净在1~200g/m L范围内线性关系良好。米卡芬净的绝对回收率为(90.1±4.4)%~(99.1±4.0)%,方法回收率为(101.7±6.0)%~(108.4±7.0)%;内标物艾瑞昔布的绝对回收率为(97.7±4.2)%。大鼠静脉注射米卡芬净15mg/kg后的主要药动学参数分别为药时曲线下面积AUC_(0→t)(307.5±33.0)g·h/m L,AUC_(0→∞)(372.2±45.7)g·h/m L,末端消除半衰期t_(1/2)(9.6±1.2)h,清除率CL(0.04±0.005)L/(h·kg),表观分布容积V(0.56±0.1)L/kg。结论本方法快速,精密,准确测定米卡芬净,可用于大鼠给药后米卡芬净的测定。
Objective An HPLC method was established for the determination of micafungin in rat plasma, and its pharmacokinetics of peramivir in rats was also investigated. Methods A Diamonsil C18 column was used, with the mobile phase of acetonitrile: 10 mmol/L ammonium acetate solution(42.5:57.5, V/V), at the detection wave length of 269 nm. The method was validated for specificity, precision and accuracy, recovery as well as stability. Results It was linear in the range of 1.0-200.0μg/mL. The extraction recoveries for micafungin were(90.1+4.4)%^(99.1+4.0)%, while its method recovery was(101.7+6.0)%-(108.4+7.0)%. The extraction recovery for the internal standard imrecoxib was(97.7+4.2)%. The major pharmacokinetic parameters after intravenous administration of micafungin 15mg/kg to rats were as follows: AUC0→t(307.5±33.0)g·h/mL,AUC0→∞(372.2±45.7)g·h/mL,t1/2(9.6±1.2)h,CL(0.04±0.005)L/(h·kg),V(0.56±0.1)L/kg. Conclusion The analytical method is quick, precise and accuracy and could be used in the analysis of concentration of micafungin in rat.
