摘要
〔目的〕探索热疗与热化疗对骨肉瘤原代细胞系耐药细胞系的影响。〔方法〕MTT法检测不同温度下热疗对原代细胞系和耐药细胞系生存率的影响;并测定43℃温度作用下浓度为1μg/m L、5μg/m L、10μg/m L的甲氨蝶呤(MTX)和浓度为1μg/m L、5μg/m L、10μg/m L的阿霉素(ADM)对原代细胞系与耐药细胞系生存率的影响;免疫组化检测43℃热疗作用下,原代细胞系与耐药细胞系HSP70的表达。〔结果〕原代细胞系与耐药细胞系热疗后细胞生存率下降;随着化疗药物浓度增加,细胞系生存率下降;热疗作用下HSP70在原代细胞系和耐药细胞系均表达增加。〔结论〕热疗可能是克服骨肉瘤细胞系耐药的一种途径。HSP 70在热处理后,原代细胞系和耐药细胞系均增加,两种细胞系HSP70表达未见差异。
〔O bjective 〕The antiosteosarcoma effects of hyperthermia were explored on primary osteosarcoma cell lines and drugresistant cell lines. 〔M ethods〕Using different temperatures and different concentrations of methotrexate( 1 μg/mL,5 μg/mL, 10 μg/mL) and doxorubicin( 1 μg/mL, 5 μg/mL, 10 μg/mL), the survival rate of primary cell lines and drugresistant cell lines HSP70 were determined by MTT assay.〔R esults〕The cell survival rate of primary cell line and drug resistant cell line was decreased after thermal therapy. The expression of HSP70 was increased in primary osteosarcoma cell line and drug resistant cell line under hyperthermia.〔C onclusion〕Hyperthermia is an effective strategy to treat the resistance of osteosarcoma cell lines.
