摘要
目的探讨MDA-7/IL-24基因联合阿霉素对肝癌模型裸鼠金属基质蛋白酶及血管内皮生长因子(vascular endothelial growth factor,VEGF)的影响。方法建立肝癌移植瘤模型裸鼠后分为模型组(A组)、阿霉素组(B组)、MDA-7/IL-24组(C组)、联合干预组(D组),并给予不同条件干预。干预后2周、6周时,测定肿瘤体积和重量、基质金属蛋白酶(MMP)、VEGF表达量。结果干预2周、6周时,B、C、D组小鼠肿瘤体积和重量均明显低于A组(P<0.05),D组小鼠肿瘤的体积和重量显著低于B组和C组(P<0.05),B、C、D组小鼠肿瘤组织中MMP3、MMP mRNA含量明显低于A组(P<0.05),D组小鼠肿瘤组织中MMP3、MMP9mRNA含量显著低于B组和C组(P<0.05),B、C、D组小鼠肿瘤组织中VEGF-C、VEGFR2 mRNA含量明显低于A组(P<0.05),D组小鼠肿瘤组织中VEGF-C、VEGFR2 mRNA含量显著低于B组和C组(P<0.05)。结论 MDA-7/IL-24基因联合阿霉素能够有效地抑制肝癌模型裸鼠的肿瘤生长,减少MMP和VEGF的表达量。
Objective To investigate the effect of MDA-7/IL-24 gene combined with adriamycin on matrix metallo- proteinase and vascular endothelial growth factor (VEGF) of hepatocarcinoma model nude mice. Methods The model nude mice with transplanted hepatocellular carcinoma were constructed and divided into model group ( group A) , Adria- mycin group (group B), MDA-7/IL-24 group (group C) and combined intervention group (group D). The groups were given different interventions. At 2 weeks and 6 weeks after the interventions, the gross tumor volume, weight, the ex- pression quantities of matrix metalloproteinases (MMP) and VEGF were measured. Results At 2 weeks and 6 weeks af- ter the interventions, the gross tumor volume and weight of mice in group B, C and D were significantly lower than those in group A (P 〈 0.05), the gross tumor volume and weight of mice in group D were significantly lower than those in group B and group C (P 〈 0.05) , the mRNA levels of MMP3 and MMP9 in tumor tissues of mice in group B, C and D were significantly lower than those in group A (P 〈 0.05) , the mRNA levels of MMP3 and MMP9 in tumor tissues of mice in group D were significantly lower than those in group B and group C (P 〈 0.05) , the mRNA levels of VEGF-C and VEGFR2 in tumor tissues of mice in group B, C and D were significantly lower than those in group A (P 〈 0.05) , and the mRNA levels of VEGF-C and VEGFR2 in tumor tissues of mice in group D were significantly lower than those in group B and group C (P 〈 0.05). Conclusion MDA-7/IL-24 gene in combination with Adriamycin can effectively in- hibit the tumor growth and reduce the expression quantities of MMP and VEGF in hepatocarcinoma model nude mice.
