摘要
目的探讨螯合Cy5-AMT-SPIONs双模探针在氯化锂-皮罗卡品诱导急性颞叶癫痫模型病灶中的靶向定位作用。方法将30只急性颞叶癫痫大鼠随机平均分为超顺氧化铁磁性纳米粒子组(SPIONs组)、Cy5标记的超顺氧化铁磁性纳米粒子组(Cy5-SPIONs组)、Cy5标记的α-甲基色氨酸的超顺氧化铁磁性纳米粒子组(Cy5-AMT-SPIONs组)。各组分别于注射药物前、注射后4 h行同参数MR T_2序列和T_2map序列并使用T_2mapping软件测量T_2值。行普鲁士蓝染色观察铁粒子在海马区的分布,在激光共聚焦显微镜下观察Cy5在大脑中的分布,行免疫荧光染色观察星形胶质细胞增生情况,行尼氏染色观察神经元缺失情况。结果 SPIONs组T_2信号稍下降,Cy5-SPIONs组T_2信号稍下降,Cy5-AMT-SPIONs组T_2信号明显下降。普鲁士蓝染色观察铁离子在海马区的分布,可见蓝棕色颗粒分布,主要分布在细胞浆内,Cy5-AMT-SPIONs组相对其他两组有明显差异。激光共聚焦显微镜下观察Cy5荧光分布各组间有明显差异。免疫荧光法各组均可见星形胶质细胞增生,各组无明显差异。尼氏染色各组均可见神经元缺失情况,各组无明显差异。结论 Cy5-AMT-SPIONs双模探针对癫痫有精确靶向定位作用且对脑组织无明显毒害作用。
Objective To investigate the target location by the action of chelating Cy 5 labeled alpha-methyl-L-trypto-phan superparamagnetic iron oxide nanoparticles dual-mode probe on lithium chloride-Piluoka induced acute temporal lobe ep-ilepcy model .Methods Thirty temporal lobe epilepsy rats were randomly and evenly assigned to superparamagnetic iron ox -ide nanoparticles group ,Cy5 labeled superparamagnetic iron oxide nanoparticles group ,Cy5 labeled alpha methyl tryptophan superparamagnetic iron oxide nanoparticles group .Each group was prior to drug injection ,injection after 4 h,with parameters MR T2 weighted images and T2 map sequence and we used T2 mapping software to measure the T2 value.Perl,iron stain was used to observe the iron distribution in the hippocampus .We used laser scanning confocal microscope to observe the distribu-tion of Cy5 in the brain.Immunofluorescence staining was used to observe the astrocyte proliferation ,Nissl staining was used to observe the neuronal loss situation .Results T2 signal of the superparamagnetic iron oxide nanoparticles group decreased slightly,T2 signal of Cy5 labeled sup superparamagnetic iron oxide nanoparticles group decreased slightly ,Cy5 labeled alpha methyl tryptophan superparamagnetic iron oxide nanoparticles group decreased significantly .The iron was mainly distributed in the cytoplasm.The Cy5-AMT-SPIONs group was significantly different from the other groups in the distribution of iron in the hippocampus area by Prussian blue staining .Laser scanning confocal microscope observation groups Cy 5 fluorescence distribu-tion had obvious differences .Immunofluorescence observed were visible astrogliosis .There was no significant difference be-tween the groups .Nissl staining of groups were visible neuron loss groups .There was no significant difference between the groups.Conclusion Cy5 labeled alpha methyl tryptophan superparamagnetic iron oxide nanoparticles dual -mode probe had an impact on precise targeting positioning function of epilepsy and had no obvious toxic effect on the brain tissue .
