康莱特注射液对人胰腺癌细胞株PANC-1增殖和凋亡影响的实验研究 被引量:4

摘要 目的通过康莱特注射液作用于人胰腺癌细胞株PANC-1,观察其对PANC-1细胞增殖和凋亡的影响,并进一步探讨其作用机制。方法倒置显微镜及电镜观察80μL·m L^(-1)康莱特注射液作用PANC-1细胞72 h后细胞形态改变;采用MTT比色法检测不同浓度(10、20、40、60、80、100μL·m L^(-1))康莱特注射液对PANC-1细胞生长增殖抑制作用的影响;同时采用流式细胞术检测不同浓度(10、20、40、60、80、100μL·m L^(-1))康莱特注射液对PANC-1细胞周期的影响。结果细胞形态学检查发现80μL·m L^(-1)康莱特注射液处理PANC-1细胞72 h后细胞生长稀疏,细胞质透亮度下降,线粒体等细胞器功能障碍,细胞核功能上受到一定的抑制;不同浓度康莱特注射液作用于PANC-1细胞株不同时段(24、48、72 h)后肿瘤细胞生长受到了一定的抑制且呈现时间和剂量依赖性;不同浓度康莱特注射液处理PANC-1胰腺癌细胞72 h后PANC-1细胞周期均阻滞于G2/M期,康莱特注射液可剂量依赖性的诱导PANC-1细胞凋亡。结论康莱特注射液能够有效抑制人胰腺癌细胞株PANC-1在体外的生长,其机制可能是通过抑制细胞增殖,促进肿瘤细胞的凋亡而实现,有望成为胰腺癌的有效治疗药物。 Objective To investigate the effects of kanglaite injection on the proliferation and apoptosis of pancreatic cancer PANC-1 cells. Methods Different concentration of sodium kanglaite injection( 10,20,40,60,80,100μL·m L^(-1)) were adder to PANC-1 cells for 24,48,72 h. Inverted microscope and electron microscope were used to observe the change of cell morphology of 80 μL·m L^(-1)kanglaite injection of PANC-1 cells after 72 h; the MTT assay was performed to reveal the inhibitory effect on cell proliferation; cell cycle was tested by flow cytometry. Results Cell morphology examination found cell growth and sparse cytoplasm brightness decreased,mitochondria dysfunction,nuclear function was inhibited,after 80 μL·m L^(-1)kanglaite injection treatment of PANC-1 cells of 72 h;kanglaite injection with different concentration treatment inhibited the proliferation in a dose and time-dependent manner; different concentration of kanglaite injection cocultured PANC-1 cells after 72 h,most of PANC-1 cell cycle arrested in G2/M phase,kanglaite injection could induce the apoptosis of PANC-1 cells dose dependently. Conclusion Kanglaite injection can effectively inhibit the growth of the human pancreatic cancer PANC-1 cells in vitro,and its mechanism may be through inhibition of cell proliferation,promote apoptosis of tumor cells and implementation,is expected to become the effective drugs for the treatment of pancreatic carcinoma.
