摘要
目的:探讨胶原诱导性关节炎(CIA)大鼠椎骨和牙槽骨的变化及泼尼松的影响。方法:58只SD雌性大鼠,随机选取8只作为正常对照组,其余用于建立CIA模型。免疫4周后,筛选出成功建立CIA大鼠并随机分成CIA模型组、泼尼松低、中、高剂量组。正常对照组和CIA模型组灌胃溶剂对照溶液,泼尼松治疗组分别灌胃泼尼松0.9、2.7、8.1 mg/(kg·d)。连续给药4个月后处死,取腰椎及上颌骨进行micro-CT扫描。结果:与正常对照组比较,CIA模型组腰椎的骨微结构明显受损和骨量明显减少,而牙槽骨的骨微结构有所破坏,但骨量变化差异无统计学意义。与CIA模型组比较,泼尼松3个剂量组腰椎和牙槽骨的骨微结构明显改善,骨量明显增加。结论:CIA大鼠椎骨丢失比牙槽骨明显。泼尼松治疗可延缓CIA大鼠椎骨和牙槽骨的骨丢失。
Objective To explore the change of vertebra and alveolar bone in collagen-induced arthritis (CIA) rats and the effects of prednisone treatment. Methods 58 female Sprague-Dawley rats were randomly divided into 2 groups, normal group with 8 rats and the remaining 50 rats were used to establish the CIA model. 4 weeks after immunization, 32 rats which were successfully infected with arthritis and randomly divided into CIA group, prednisone treatment groups.Rats in control group were received vehicle via gavage as well as CIA group, while rats in the other three groups were treated with prednisone at 0.9 mg·kg^-1·d^-1, 2.7 mg·kg^-1·d^-1 or 8.1 mg·kg^-1·d^-1. After 4 months of administration, all rats were sacrificed, and the maxillary and lumbar were collected for micro-CT scanning to observe the changes of bone microstructure. Results Compared with control group, destruction of bone microstructure and bone loss in lumbar was obviously seen in CIA group. Microstructure of alveolar bone was damaged, but there was no significant difference in bone mass between CIA group and control group. Compared with CIA group, bone microstructure was improved and bone mass of lumbar and alveolar bone was increased in all groups treated with prednisone. Conclusion Bone loss of vertebra was more serious than that of alveolar bone in CIA rats. Prednisone treatment could prevent bone loss of vertebra and alveolar bone in CIA rats.
