miR-586在骨肉瘤患者体内的表达及其意义

摘要 目的:探讨miR-586在骨肉瘤患者体内的表达水平及临床意义。方法:以20例骨肉瘤患者及20例健康体检者作为研究对象,收集其静脉血、瘤体组织及瘤体周围正常骨组织,提取总RNA,采用实时荧光定量PCR检测骨肉瘤患者与健康志愿者各样本中miR-586的表达水平。评估血清miR-586表达水平与瘤体内表达水平的相关性及miR-586的诊断效能。结果:骨肉瘤患者血清中miR-586表达水平较健康对照组高(P<0.05),瘤体组织中miR-586相对表达水平明显高于瘤体周围正常骨组织(P<0.05),骨肉瘤患者瘤体组织内miR-586表达水平与血清中表达水平呈正相关(r=0.908,P<0.001)。ROC曲线下面积为0.883,当最佳筛查阳性界值为0.9550时,敏感性和特异性均为85%。结论:患者血清中miR-586或可作为骨肉瘤诊断的一个血清学指标。
