摘要
目的:建立UPLC法同时测定小叶榕不同部位的样品中表阿夫儿茶精、牡荆素、异牡荆苷的含量。方法:采用Acquity UPLC BEH C_(18)色谱柱(2.1 mm×100 mm,1.7μm),以甲醇-0.1%磷酸水溶液为流动相,梯度洗脱流速0.3 mL·min^(-1),检测波长270 nm,柱温35℃,进样量3μL。结果:表阿夫儿茶精、牡荆素、异牡荆苷3个成分的质量浓度分别在0.002 8~0.028 0、0.005 04~0.050 4和0.009 28~0.092 8mg·mL^(-1)范围内呈良好线性关系(r=0.999 9),平均加样回收率(n=9)分别为98.2%、98.8%和99.1%。小叶榕叶中同时检测出表阿夫儿茶精、牡荆素、异牡荆苷,果实中检测出牡荆素、异牡荆苷,而茎和气生根中仅检测出牡荆素,4个部位的样品中上述3个成分的含量范围分别为0-0.177、0.081~0.509、0-0.509 mg·g^(-1)。结论:所建立的方法经方法学验证,可用于小叶榕药材的质量控制和评价。小叶榕叶中同时检测出3个成分,且其中2个成分含量明显高于其他部位,与传统上以叶入药相符。
Objective: To develop a method for simultaneous determination of the amounts of three flavonoids ( ( - ) - epiafzelechin, vitexin, isovitexin )in different parts of Ficus microcarpa L.f. by ultra-high-performance liquid chromatography ( UPLC ). Methods: UPLC was performed on a Waters Acquity UPLC BEH-C18 analytical column ( 2.1 mm × 100 mm, 1.7 μm )at 35 ℃. Methanol and 0.1% phosphoric acid was adopted as the mobile phase by gradient elution at the flow rate of 0.3 mL·min-1. The detector wavelength was set at 270 nm and the injection volume was 3 μL. Results: Good linearity was shown in the range of 0.002 8-0.028 0, 0.005 04-0.050 4 and 0.009 28- 0.092 8 mg· mL-1 of( - )-epiafzelechin, vitexin and isovitexin, respectively ( r ≥ 0.999 9 ), and the average recoveries ( n=9 )were 98.2%, 98.8% and 99. I%, respectively. ( - )-Epiafzelechin, vitexin, isovitexin weredetected in the leaf; vitexin and isovitexin were detected in the fruit; while only vitexin was detected in the stem and aerial root. The content ranges were 0-0.177, 0.081-0.509 and 0-0.509 mg·g-1.Conclusion: The established method is proved to be a reliable approach for the quality control and quantity evaluation of Ficus microcarpa L. f.. The leaf contained all these three constituents, two of which were of higher amount than those in other parts. Our findings were in accordance with the traditional therapy which selected the leaf as the medicinal part of this plant.
