摘要
[目的]木葡聚糖内转糖苷酶/水解酶(XTH)属于糖苷水解酶GH16家族,其家族成员可能在植物响应逆境胁迫的过程中发挥重要作用,为深入挖掘谷子抗旱基因,进而选育谷子抗逆新品种,[方法]本研究利用生物信息学手段,对谷子XTH基因家族进行了分析。[结果]从谷子基因组数据库中鉴定出16个XTH基因。结构分析表明:SiXTH家族成员在基因结构及编码区序列上较为保守,含1~3个内含子;谷子XTH蛋白含有XTH家族典型的保守基序DEIDFEFLG;预测SiXTH基因家族成员在启动子区域含有响应逆境胁迫的顺式作用元件。在干旱胁迫下,通过比较两个谷子品种勾勾母鸡咀及晋汾16在干旱胁迫下基因的相对表达水平,我们发现了3个上调表达和1个下调表达的SiXTH基因。[结论]因此推测,同一类基因其基因结构及蛋白结构域相似,且XTH基因家族在谷子应答干旱胁迫的过程中起一定的作用,同时本研究也为深入探究XTH基因家族成员的功能奠定了一定的基础。
[Objective] Xyloglucan endotransglycosylase/hydrolase(XTH) is in the family of 16 glycoside hydrolases, its members may play an important role in plant stress response. In order to explore drought genetic resources and breed the new varieties of foxtail millet with high resistance, [Methods]in this study, the SIXTH gene family was investiga- ted according to the foxtail millet genome database and bioinformatic method. [Results] A total of 16 XTH genes were identified. Gene structure analysis showed that the coding sequences and structure of the SIXTH genes were very con- servative, the number of introns among these genes ranged from 1 to 3 ; SiXTHs had the typical conservative DEI- DFEFLG motif; SiXTHs have stress related cis-acting elements in promoter regions. Analysis of the expression of SiXTHs under drought stress in foxtail millet varieties GG and JF16, showed that three SIXTH genes was up-regula- ted, but one gene down-regulated. [Conclusion]We assumed that the genes in the same subgroup had the same intron- exon structure and domain. SIXTH genes might play an important role in foxtail millet under drought stress. This study would be useful for further analysis of the function of the gene family members.
