摘要
目的明确微型核糖核酸-29 c(microRNA-29 c)与血清和糖皮质激素诱导蛋白激酶1(serum and glucocorticoid-inducible kinases 1,SGK 1)的靶向调控关系,并探讨该调控关系与子宫内膜异位症伴不孕的关系。方法收集2015年1月至2016年3月于德州市中医院行腹腔镜手术治疗的单纯子宫内膜异位症患者(24例)、子宫内膜异位症伴不孕患者(22例)分泌期在位子宫内膜上皮细胞,另取同期健康对象(18例)的分泌期在位子宫内膜上皮细胞,采用荧光定量聚合酶链反应技术(quantitative polymerase chain reaction,q PCR)检测并对比3组miRNA-29 c及SGK 1 mRNA表达量,分析miRNA-29 c与SGK 1 mRNA相关性;以子宫内膜异位症伴不孕患者子宫内膜细胞为标本,原代培养人子宫内膜细胞,以pre-miRNA-29 c转染,使miRNA-29 c在细胞中过表达,应用q PCR技术检测miRNA-29 c与SGK 1 mRNA变化情况。结果 1子宫内膜异位症伴不孕患者miRNA-29 c及SGK 1 mRNA表达量明显高于单纯子宫内膜异位症患者及正常对照组(P<0.05);miRNA-29 c与SGK 1 mRNA的表达呈显著正相关(P<0.05);2 miRNA-29 c过表达后,SGK 1 mRNA的表达量明显上升(P<0.05)。结论 miRNA-29 c能够正向调控SGK1 mRNA的表达,这可能是导致子宫内膜异位症患者不孕的机制之一。
Objective To define the targeting regulation relation of miRNA - 29c to serum and glucocorticoid - inducible kinases 1 ( SGK1 ) , and to explore the relationship between the targeting regulation and endometriosis infertility. Methods From January 2015 to March 2016, endometrial epithelial cells samples from 24 cases of endometriosis patients and 22 cases of endometriosis infertility patients who underwent laparoscopic surgery, and 18 cases of healthy women in Traditional Chinese Medicine Hospital of Dezhou were collected. The expressions of miRNA -29c and SGK1 mRNA were detected by quantitative polymerase chain reaction(qPCR) , and thecorrelation between miRNA -29c and SGK1 mRNA was analyzed. Tissue samples from endometriosis infertility patients were primary cultured and transfected by Pre - miRNA - 29c. The changes of miRNA - 29c and SGK1 mRNA were detected by qPCR. Results ① The expressions of miRNA - 29c and SGK1 mRNA in tissue samples from endometriosis infertility patients were significantly higher than those from endometriosis patients and healthy women, the differences were statistically significant ( P 〈 0. 05 ). The expressions of miRNA - 29c and SGK1 mRNA showed a significant positive correlation (P 〈 0. 05 ) ; ② With the overexpression of miRNA - 29c, the expression of SGK1 mRNA significantly increased (P 〈 0. 05). Conclusion miRNA - 29c can positively regulate the expression of SGKt mRNA, which may be one of the mechanisms leading to infertility in patients with endometriosis.
