摘要
目的探讨细胞因子信号传导负调控因子1(SOCS1)在妊娠期肝内胆汁淤积症(ICP)母胎界面的表达以及意义。方法采用ELISA的方法检测18例ICP孕妇(ICP组)与20例晚孕孕妇(对照组)的胎盘组织匀浆IL-4和TNF-α浓度,采用免疫组化法测定两组胎盘组织中SOCS1的定位与表达水平。结果 1SOCS1主要表达于滋养细胞。2ICP组SOCS1阳性细胞率显著低于对照组,差异有统计学意义(P<0.01)。3ICP组的IL-4表达水平为(1.08±0.15)ng/L,显著低于对照组的(1.56±0.32)ng/L,差异有统计学意义(P<0.01);ICP组的TNF-α表达水平为(18.37±1.68)ng/L,显著高于对照组的(10.59±1.63)ng/L,差异有统计学意义(P<0.01)。4ICP组的TNF-α/IL-4比值为17.01±2.06,显著高于对照组的6.78±0.54,差异有统计学意义(P<0.01)。结论 SOCS1表达水平明显降低,可能参与了ICP的发病,这可能与Th1/Th2比例失衡有关。
Objective To explore the expression and significance of suppressor of cytokine singnaling-1 (SOCS 1) in the placenta ofintrahepatic cholestasis during pregnancy (ICP). Methods The samples were obtained from 18 cases of ICP pregnant women (ICP group)and 20 cases of normal later pregnant women (control group), and the levels of IL-4 and TNF-a of placenta homogenate were detected byELISA. The expression level and localization of SOCS1 in the placenta were examined by immunohistochemical method. Results TheSOCS 1 was mainly expressed in the chorion cells. (The SOCS 1 positive cell rate of ICP group was significantly lower than that of controlgroup, with statistical difference (P 〈0.01). The expression level of IL-4 of ICP group was (1.08 ± 0.15) ng/L, significantly lower than(1.56 ± 0.32) ng/L of control group, with statistical difference (P 〈0.01). The TNF-a level of ICP group was (18.37 ± 1.68) ng/L,significantly higher than (10.59 ± 1.63) ng/L of control group, with statistical difference (P〈0.01). The ratio of TNF-a/IL-4 of ICP groupwas 17.01 ± 2.06, significantly higher than 6.78 ± 0.54 of control group, with statistical difference (P〈0.01). Conclusions The expressionlevel of SOCS1 significantly decreases, which means that SOCS1 probably plays a role in the pathogenesis of ICP and is relative with thedisequilibrium of Th 1/Th2.
