丹红注射液对动脉粥样硬化家兔内皮功能及肝X受体mRNA的影响 被引量:8

摘要 【目的】观察丹红注射液对动脉粥样硬化(AS)模型家兔内皮功能及肝X受体(LXR)mRNA表达的影响。【方法】将60只新西兰雄性白兔随机均分为正常对照组、模型组、丹红注射液组;除正常对照组外,模型组和丹红注射液组家兔均采用喂饲高胆固醇饲料联合注射牛血清白蛋白的方法复制AS模型,丹红注射液组给予肌注丹红注射液(0.3 m L·kg^(-1)·d^(-1))。在第1周和第8周取材,采血并分离肝脏和左髂外动脉。运用酶法检测各组家兔血浆一氧化氮(NO)、血管性血友病因子(v WF)、内皮素-1(ET-1);实时荧光定量聚合酶链反应法检测各组家兔肝脏肝X受体α(LXRα)mRNA和肝X受体β(LXRβ)mRNA;组织形态学分析各组家兔左侧髂外动脉AS斑块形成情况。【结果】第1周时,3组家兔血浆NO、v WF、ET-1水平及LXR mRNA表达比较,差异无统计学意义(P>0.05)。第8周时,与正常对照组比较,模型组血浆NO水平显著降低(P<0.01),v WF、ET-1水平显著升高(P<0.01),LXRαmRNA表达显著升高(P<0.01),LXRβmRNA表达差异无统计学意义(P>0.05),血管AS病变明显;与模型组比较,丹红注射液组血浆NO水平显著升高(P<0.01),v WF、ET-1水平显著降低(P<0.01),LXRαmRNA表达显著降低(P<0.01),LXRβmRNA表达差异无统计学意义(P>0.05),血管AS病变明显减轻。【结论】丹红注射液能够改善AS家兔的血管内皮功能,延缓AS板块的进展,其机制可能与调控肝脏LXRα表达的作用有关。 Objective To observe the effect of Danhong Injection on the vascular endothelial function and liver X receptor(LXR)mRNA in rabbit model of experimental atherosclerosis,and to explore its possible mechanism.Methods Sixty New Zealand male rabbits were randomly divided into normal control group, model group, and Danhong Injection group(Danhong Injection in the dosage of 0.3 m L·kg^(-1)·d^(-1)). Expect for the normal control group, the other two groups were fed with high-fat diet and given injection of bovine serum albumin to induce atherosclerosis model. On week one and week 8,the blood,liver,left external iliac artery were sampled for the experiment. Plasma nitric oxide(NO),von willebrand factor(v WF),and endothelin-1(ET-1) were detected by enzymatic assay. LXRα mRNA and LXRβ mRNA in rabbit liver of various groups were detected by real-time fluorescence quantitative polymerase chain reaction. The plaque formation in left iliac artery was observed by histomorphological analysis method. Results On week one, there were no significant differences of the serum NO,v WF,ET-1 and LXR mRNA between the three groups(P 0.05). On week 8,compared with the normal control group,the plasma NO level was decreased(P 0.01),the v WF,ET-1 and LXRα mRNA levels were significantly increased(P 0.01),the LXRβ mRNA level had no significant changes(P 0.05),and the plaque size of left iliac artery was enlarged in the model group. Compared with the model group,the plasma NO level was increased(P 0.01),the v WF,ET-1 and LXRα mRNA levels were significantly decreased(P 0.01),the LXRβ mRNA level had no significant changes(P 0.05),and the plaque size of left iliac artery was reduced(P 0.01) in Danhong Injection group. Conclusion Danhong injection can improve endothelial function and delay the development of atherosclerosis plaque of experimental atherosclerosis rabbits,which may be related with the regulation of LXRα expression.
