摘要
目的检测miR-150-5p在肝细胞癌组织及癌旁组织中的表达,分析miR-150-5p表达水平与肝细胞癌患者临床病理特征和术后5年总体生存率的关系。方法利用实时定量聚合酶链反应(qRT-PCR)和原位杂交方法检测91例肝细胞癌组织及配对癌旁组织中miR-150-5P的表达情况,并进行统计分析。结果qRT-PCR结果显示miR·150-5p在肝细胞癌组织中表达水平显著低于癌旁组织,原位杂交结果显示肝细胞癌组织中miR-150-5P阳性率显著低于癌旁组织(χ2=4.958,P=0.046)。χ2检验结果显示肝细胞癌组织中miR-150-5P的表达水平与肝细胞癌患者就诊时淋巴转移情况有关(χ2=8.617,P=0.006),而与其他临床病理特征之间无关。生存曲线显示miR-150-5P表达水平与肝细胞癌患者术后5年总体生存率密切相关(χ2=6.184,P=0.013)。结论miR-150-5P在肝细胞癌组织中表达降低,其表达与肝细胞癌患者淋巴转移情况有关,亦与肝细胞癌患者术后5年总生存率有关。
Objective To explore the expression and clinical significance of miR-150-5P in patients with hepatocellular carcinoma (HCC). Methods Expressions of miR-150-SP were analyzed by quantita- tive real-time polymerase chain reaction (qRT-PCR) and in situ hybridization (ISH) in cancerous and ad- jacent tissues of 91 HCC patients. Statistical analysis was conducted by SPSS 22. 0 software. Results qRT-PCR revealed that the mRNA expression level of miR-150-5P was significantly lower in HCC tissues than adjacent tissues. ISH revealed that the positive rate of miR-150-5P was downregnlated in HCC tissues compared to adjacent tissues(χ2 =4. 958, P =0. 046). Chi-square analysis revealed that high miR-150-5P expression was inversely associated with lymph node status of HCC patients, which was statistically significant (χ2 = 8.617,P = 0. 006 ). Kaplan-Meier curves revealed that high miR-150-5P expression was positivelycorrelated with 5-year overall survival rates of HCC patients after surgery, which was also statistically sig- nifieant(χ2 =6. 184,P=0.013). Condusions miR-150-5P expression is downregulated in HCC, and is negatively correlated with lymph node metastasis and positively correlated with cumulative survival of HCC patients.
