摘要
目的观察毛蕊异黄酮对顺铂抑制胃癌细胞增殖的增强作用。方法将BGC823细胞分为4组,分别用生理盐水、顺铂、毛蕊异黄酮和2种药物联合处理,检测不同时间点、不同处理因素下的细胞增殖情况。进一步检测影响细胞增殖的相关蛋白,确定毛蕊异黄酮联合顺铂抑制胃癌细胞增殖的方式。结果对细胞作用24 h后,20μg/m L顺铂,10μg/m L毛蕊异黄酮,以及2种药物联合处理组对BGC823细胞的增殖抑制率分别为56.44%±2.08%,9.52%±2.77%和74.44%±0.82%。药物联合使用后对细胞的抑制率明显高于单个药物处理组,各组数据间差异有统计学意义(P<0.05)。并且无论是在蛋白水平还是RNA水平上,毛蕊异黄酮均可以显著增强顺铂对Cyclin D1,CDK4和CDK6蛋白的抑制作用。结论毛蕊异黄酮可以显著提高顺铂对BGC823细胞增殖的抑制率,2种药物联合使用可以降低顺铂单独使用所导致的毒副作用。
Objective To observe the effect of calycosin on cisplatin-induced inhibition of human gastric BGC823 ceils. Methods BGC823 ceils were treated with different drugs : saline, cisplafin, ealycosin and cisplatin combined with calycosin. MTY assays were used to detect the prolif- eration rate of BGC823 cell. Then the protein level and RNA level of cychn D1, CDK4 and CDK6 were detected by Western blotting and Real- time PCR. Results The proliferation inhibition rates of BGC823 cells lxeated with 20 p.g/mL cisplatin, 10μg/mL meperoflavone, and combina- tion of the 2 drugs were 56.44%± 2.08%, 9.52%±2.77% and 74.44%±0.82%, respectively. The inhibition rate of the combination of drugs was sigrfificantly higher than that of the single drug treatment group (P 〈 0.05). In addition, we found that ealyeosin can significantly enhance the inhi- bition of Cyclin D 1, CDK4 and CDK6 by cisplatin in protein level and RNA level. Conclusion Calycosin can significantly increase the inhibitory rate of cisplatin on BGC823 ceil proliferation, and the combination of the two drugs can reduce the side effects of cisplatin.
