摘要
采用乙醇浸提法提取胡椒中的胡椒碱,通过单因素和L_(16)(4~5)正交实验,确定了最佳提取工艺条件:提取温度为65℃,乙醇浓度为75%,料液比为1∶30,提取时间为0.5h,提取次数为3次。通过静态和动态实验,确定了大孔树脂分离胡椒碱的最佳工艺条件:树脂种类为AB-8,上柱浸提液浓度为1.1mg/mL,pH值为6,洗脱液为体积分数75%、pH值为5的乙醇溶液,胡椒碱纯度由浸提液的16.46%上升到69.32%。最后,通过C_(18)层析柱对大孔树脂纯化的胡椒碱粗品进一步纯化,用90%乙醇洗脱,经HPLC测定纯化后粗品中的胡椒碱纯度上升为81.26%。
The piperine is extracted from pepper with ethanol extraction method, and the optimal extraction conditions are determined by single factor and L16 (45) orthogonal experiments, the ethanol concentration of 75%, the extraction temperature of 65℃, the solid-liquid ratio of 1:30, the extraction time of 0.5, the extraction times of 3.
