摘要
【目的】探讨年龄相关听力损失小鼠耳蜗组织中甲基化CrG结合蛋白2(MeCP2)的表达差异及临床意义。【方法】在对不同月龄BALB/c小鼠听功能和耳蝎形态学研究以及耳蜗MeCP2蛋白表达免疫组化研究的基础上,应用半定量反转录多聚酶链反应方法,检测3、R、12、18个月月龄鼠耳蜗组织中McP2基因mRNA的表达。【结果】MeCP2在4组不同月龄小鼠耳蜗组织中均确表达,MeCP2mRNA表达水平随着月龄增加逐渐下降,冬锕闻萼辟确缔砂哔意义.(P〈0.01)。【结论】MeCP2在年龄相关听力损失小鼠耳蜗组织中的表达水平随着小轧月龄的增加而降低,这可能与耳蜗的老化及老年性耳聋的发生有关。
[Objective]To investigate the variance of the methyl-CpG-binding protein 2 (MeCP2) expression in the cochlea of BALB/c mice with age-related hearing loss and its clinical significance. [Methods]On the basis of research on auditory function of BALB/c mice in different months, the morphological study of cochlea and immunohistochemieal ext, reSsion ot MeCP2 protein in cochlea, the expression of MeCP2 mRNA gene was detected in 3, 6, 12, 18 months old rats in the cochlea through the method of semi quantitative reverse transcription polymerase chain reaction. [Results] MeCP2 was expressed in 4 mice groups in the cochlea of BALB/c with different months of oge, the expres,qion level of MeCP2 mRNA decreased gradually with the increase of month age, the difference was statistically significant ( P〈 0.01).[Conclusion] The expression level of MeCP2 in the cochlea of BALB/c mice with age-related hearing loss decreased with the increase of the age of mice, which may be related to the aging of cochlea and the occurrence of preshycusis.
