摘要
目的:探讨microRNA-296-5p(miR-296-5p)对胰腺癌细胞体外生物学行为的影响。方法:分别转染miR-296-5p类似物(Agomir)及miR-296-5p拮抗物(Antagomir)至胰腺癌细胞系(As PC-1、Panc-1和Capan-2),应用CCK-8法、Annexin-V APC/7-AAD法、细胞黏附实验、Transwell侵袭迁移实验检测miR-296-5p对胰腺癌细胞增殖、凋亡、黏附、侵袭和迁移能力的影响。结果:转染miR-296-5p Agomir后胰腺癌细胞增殖、侵袭及迁移能力显著降低,凋亡率和黏附能力增加;而转染miR-296-5p Antagomir后胰腺癌细胞增殖、侵袭及迁移能力增强,凋亡率和黏附能力降低。结论:体外实验证实miR-296-5p影响胰腺癌细胞体外生物学行为,抑制其转移。
Objective:To explore the effect of microRNA - 296 - 5p(miR - 296 - 5p)on the biological behavior of pancreatic cancer cells. Methods:The pancreatic cancer cell lines were transiently transfected with miR - 296 - 5p Agomir and miR - 296 - 5p Antagomir. Cell proliferation was evaluated using the Cell Counting Kit - 8(CCK - 8) assay and apoptosis was assessed by flow cytometry. Adhesion was evaluated by cell adhesion experiments. Invasio-nand migration were measured by Transwell chamber assays. Results:The overexpression of miR - 296 - 5p inhibited proliferation abilities as shown in the cells transfected with the miR - 296 - 5p Agomir,in additional,the miR - 296 -5p accelerated apoptosis and enhanced adhension. The overexpression of miR - 296 - 5p inhibited invasion and migra-tion abilities,as shown in the cells transfected with the miR - 296 - 5p Agomir. On the contrary,when transfecting miR - 296 - 5p Antagomir,the results were completely opposite. Conclusion:The experiment confirmed miR - 296 -5p can influence biological behavior of pancreatic cancer cells in vitro,and inhibited metastasis.
