摘要
蛋氨酸是一种必需氨基酸,在哺乳动物的蛋白质合成、DNA甲基化和多胺合成等方面起着重要作用。多数肿瘤细胞在缺乏蛋氨酸的环境下不能正常生长、增殖,为此依据蛋氨酸依赖性成为选择性治疗肿瘤的潜在方法。通过饮食限制蛋氨酸的摄入和蛋氨酸全肠外营养法能明显减慢肿瘤生长速度,降低转移率,延长生存期。体内注射蛋氨酸酶对肿瘤生长增殖的抑制作用更加显著。蛋氨酸酶不仅能单独抑制肿瘤生长,亦可增强化疗药物对肿瘤的抑制作用。通过基因工程构建不同来源的蛋氨酸酶,在大肠杆菌内高效表达纯化之后对GLC细胞有良好的生长抑制作用。重组蛋氨酸酶腺病毒对肿瘤细胞也有较强的杀伤作用。
Methionine is an essential amino acid,plays an important role in protein synthesis,DNA methylation, polyamine synthesis and other aspects in mammals. Most cancer cells can not grow and proliferation normally in the absence of methionine environment,so methionine dependence as a potential selective method for the treatment of tumors. By dietary restriction of methionine and intake of total parenteral nutrition method can significantly slow the rate of tumor growth,reducing the transfer rate and prolong animal survival. Inhibitory effect of methionine enzyme on growth and proliferation of tumor is more obvious. Methionine enzyme can not only separate inhibition of tumor growth, also enhance the inhibition of chemotherapeutic drugs on tumor. Recombinant methioninase adenovirus have a strong killing effect on tumor cells.
