红景天苷对心肌缺血-再灌注大鼠心肌梗死面积及细胞凋亡的影响 被引量:9

Influence of Salidroside on Myocardial Infarctional Size and Apoptosis Index in Rats with Myocardial Ischemia Reperfusion Injury

摘要 目的观察红景天苷对心肌缺血再灌注损伤(MIRI)大鼠心肌梗死面积及心肌细胞凋亡的影响,探讨大株红景天抗MIRI的作用。方法将SD大鼠按照随机数字表法分为6组,每组6只:假手术组(Sham)、心肌缺血-再灌注模型组(I/R)、红景天苷预防组(红景天苷+缺血再灌注,S+I/R)、红景天苷治疗组(缺血再灌注+红景天苷,I/R+S)、红景天苷预防组+磷脂酰肌醇-3-激酶(PI3K)特异性抑制剂LY294002组(S+I/R+LY组)、红景天苷治疗组+PI3K特异性抑制剂LY294002组(I/R+S+LY组)。红景天苷预防组和预防组+LY组造模前连续给药3 d;红景天苷治疗组和治疗组+LY组再灌注即刻加予红景天苷;Sham组和I/R组均给予等量生理盐水腹腔注射;预防+LY组和治疗+LY组于冠脉左前降支(LAD)结扎前30 min腹腔注射PI3K特异性抑制剂LY294002。心肌缺血30 min再灌注24 h后,迅速摘取大鼠心脏留取心室,运用Evans’blue及TTC法双重染色,确定心肌梗死区和缺血区面积;运用末端标记法(TUNEL)染色法检测心肌细胞凋亡并计算心肌细胞凋亡指数。结果与I/R组比较,红景天苷预防组和治疗组均能显著缩小MIRI大鼠的心肌梗死面积(P<0.05),TUNEL法检测阳性细胞核显著减少,差异有统计学意义(P<0.05);与预防组和治疗组比较,预防+LY组及治疗+LY组心肌梗死面积显著扩大,细胞凋亡指数显著升高,差异有统计学意义(P<0.05)。结论红景天苷能有效减少MIRI大鼠心肌梗死面积和心肌细胞凋亡指数。 Objective To observe the effect of salidroside on myocardial size and apoptosis index in rats with myocardial ischemia reperfusion injury( MIRI),to investigate the function of salidroside repel MIRI. Methods Sprague-Dawley rats were randomly divided into 6 groups :sham operation group( S group),ischemia- reperfusion group( I / R group),salidroside preventive group( first treating with salidroside and then inducing ischemia-reperfusion) and salidroside treatment group( first inducing ischemia-reperfusion and then treating with salidroside),salidroside preventive + LY294002 group,salidroside treatment + LY294002 group,6 rats in each group. Both of the salidroside preventive group and salidroside preventive + LY group administered continuously modeling three days before,salidroside treatment group and salidroside treatment + LY group in addition to the salidroside immediate reperfusion. Rats in sham operation group and ischemia- reperfusion group( I / R group) received a same volume of 0. 9% Na Cl. The salidroside preventive + LY group and treatment + LY group with an additional injection of PI3 K inhibitor( LY294002) injection slowly 35 min before ligation. Myocardial ischemia sizes were measured by Evans blue and2,3,5-triphenyltetrazolium chloride( TTC) staining. Myocardium apoptosis rate was measured by TUNEL. Results Compared with I / R group,in both salidroside preventive and treatment groups,myocardial infact size reduced and the apoptosis index lower remarkable(P〈0.05),Compared with salidroside treatment group and preventive group,preventive + LY group and treatment + LY group myocardial infact size increased,apoptosis index increased(P〈0.05). Conclusion Salidroside can reduce myocardial infarctional size and apoptosis index in rats with MIRI.
