猪圆环病毒2型吉林株的分离鉴定及全基因序列分析

Isolation and Identification of Porcine Circovirus 2(PCV2) Isolated from Jilin Province and Its Complete Genome Sequence Analysis

摘要 利用大体病理剖检、病理组织学观察、病原分离、PCR检测等方法对长春郊区一例猪圆环病毒病进行了病理学和病原学诊断,证实造成病猪死亡的原因为猪圆环病毒2型(porcine circovirus2,PCV2)感染所致,并将该分离毒株命名为PCV2-JL/China/2015。参照Genbank已公布的PCV2基因序列,设计合成一对用于PCV2全基因序列扩增的特异性引物,成功获得PCV2-JL/China/2015毒株的全基因序列。序列测定结果显示,PCV2-JL/China/2015毒株基因组全长为1 767 bp;遗传进化树分析结果显示,该分离株与我国湖南长沙病毒株FJ594471-1C01分离株亲缘关系较近,处于同一进化分支上。 An suspected porcine circovirus disease was confirmed by necropsy, histopathology observation, virus isolation and PCR detection, which was caused by porcine circovirus 2 (PCV2) infection. A porcine circovirus strain (PCV2-JL/China/2015 ) was suecessfully isolated fromspleens and lymph nodesby passaging in PK-15 ceils. A pair of primers based on PCV2 was designed according to the published sequences in GenBank. The length of complete genome of PCV2-JL/China/2015 was 1 767 bp by sequencing analysis. The results of the phylogenetic tree analysis indicated that there was a close genetic relationship between P CV2-JL/China/2015 and F J594471-1 CO 1 isolate from Changsha city.
