摘要
目的通过对不同品种、不同栽培方式及不同月龄的金线莲中游离单糖及多糖中单糖的构成做了较全面地分析,比较不同品种、栽培方式及时间对金线莲中多糖含量的影响,筛选优势品种及栽培方式,为生产企业种植金线莲及制定质量标准提供参考。方法采用PMP柱前衍生HPLC,梯度洗脱分离测定不同金线莲中7种单糖含量。结果 8个金线莲样品中的游离单糖均以葡萄糖占绝对优势。不同品种金线莲多糖的单糖构成种类并无差异,由甘露糖、鼠李糖、半乳糖醛酸、葡萄糖、半乳糖、阿拉伯糖6种单糖构成,并以葡萄糖为优势构成。福建种的多糖含量高于台湾种和福建野生种,组培金线莲随着移植至林下,多糖含量明显增高,但移植时间越长,多糖含量反而降低。结论组培5个月,移植林下4个月的福建种金线莲的多糖含量最高。本实验采取的HPLC法具有简便,重复性好、稳定性好、灵敏度高等特点,适用于金线莲中游离单糖及多糖的含量测定。
Objective To Select predominant varieties and cultural methods, we analyse free monosaccharide and polysaccharide in different varieties and different ages ofAnoectochilus roxburghii. Methods The seven monosaccharide derivatives obtained with 1-Phenyl-3-methyl-5-pyrazolone (PMP) were separated by HPLC using a developed gradient elution process. Results Monosaccharide composition in eight Anoectochilus roxburghii polysaccharide, composed of six kinds of monosaccharides: mannose, rhamnose, galacturonic acid, glucose, galactose, arabinose, were no difference, and glucose was the dominant constitute. Polysaccharide content of of Fujian were higher than Taiwan Varietiehs's, else higher than Fujian wild species's. Polysaccharide content was significantly increased with the transplantation to the forest, but transplant time longer, polysaccharide content decreased. Conclusion The highest polysaccharide content appears in Fujian Anoectochilus roxburghii when it was tissue cultured for 5 months and forest transplanted for 4 months. The method can be used to determine the content of free monosaccharides and polysaccharides in Anoectochilus roxburghii with accurate results and good repeatability.
