摘要
目的:建立一种快速检测O1群小川型霍乱弧菌的近红外免疫层析方法。方法:利用近红外免疫层析技术,采用双抗体夹心法检测O1群小川型霍乱弧菌,制备靶向于O1群小川型霍乱弧菌的免疫层析试纸条。结果:整个检测方法在45 min内即可完成,检测O1群小川型霍乱弧菌的最低检出浓度为1×10~2 cfu/mL。特异性实验表明,该试纸条与O1群小川型霍乱弧菌样品发生特异性反应,与沙门氏菌、金黄色葡萄球菌、志贺氏菌、单增李斯特菌、副溶血弧菌无交叉反应。与常用的胶体金方法相比,近红外免疫层析法对靶标菌的灵敏度提高了100倍。结论:新研制的O1群小川型霍乱弧菌试纸条具有特异性强、灵敏度高的特点,实验简单、方便,满足口岸现场快速检测的需要,提高检验检疫效率。
Objective: To establish a rapid detection of near-infrared immunochromatographic method of Vibrio cholerae O1 Ogawa. Method: Using near-infrared immunochromatography to detect Vibrio cholerae O1 Ogawa, we use double antibody sandwich method for preparing targeting Vibrio cholerae O1 Ogawa immunochromatographic test strip. Results: The results of the test strip and Vibrio cholerae O1 Ogawa sample have specific reaction and no cross-reaction with Salmonella, Staphylococcus aureus, Shigella, Listeria monocytogenes and Vibrio parahaemolyticus. This method can be completed in 45 min. The minimum detectable concentration detection of Vibrio cholerae O1 Ogawa was 1×10^2 cfu/mL. Compared with colloidal gold method, near-infrared immunochromatography sensitivity of target bacteria 100 times. Conclusion: The newly developed Vibrio cholerae O1 Ogawa test strip has strong specificity and high sensitivity. The experiment is simple and easy to meet the needs of port-site rapid test. This will improve the efficiency of inspection and quarantine.
