摘要
目的探讨Neuritin蛋白在非小细胞肺癌(NSCLC)组织中的表达及其与临床病理特征和预后的相关性。方法收集在该院行手术切除的87例NSCLC组织及其对应的87例癌旁组织,采用免疫组织化学法检测癌及癌旁组织中神经突起生长因子(Neuritin)蛋白的表达,并根据二级计分法将其分为低表达和高表达部分,分析Neuritin蛋白表达水平与临床病理参数[病理类型、分化程度、肿瘤大小、淋巴结有无转移、表皮生长因子受体(EGFR)表达等]的相关性。结果 Neuritin蛋白的阳性表达位于细胞质,NSCLC组织中Neuritin蛋白以高表达为主,其表达率为75.86%(66/87),远高于癌旁组织的31.03%(27/87),差异有统计学意义(P<0.05);且NSCLC组织中Neuritin蛋白表达水平与NSCLC病理类型、分化程度、肿瘤大小、EGFR基因突变相关(P<0.05),与年龄、性别、吸烟与否、肿瘤部位、有无淋巴结转移无关(P>0.05);Neuritin高表达组的中位疾病无进展生存期(PFS)为16个月,低表达组的PFS为13个月,差异无统计学意义(P>0.05);随访数据中Neuritin高表达组的中位总生存期为37个月,低表达组为47个月,差异无统计学意义(P>0.05);结论 Neuritin蛋白在NSCLC及其癌旁组织中均有表达,其中在NSCLC组织中呈高表达,且与NSCLC病理类型、分化程度、肿瘤大小、EGFR基因突变相关,提示Neuritin可能在NSCLC的发生、发展中发挥一定作用。
Objective To investigate the expression status of neuritin protein and its association with clinical pathological features and prognosis in non-small cell lung carcinoma (NSCLC). Methods NSCLC tissues and the corresponding para-cancerous tissues were collected from 87 cases in our hospital. SP immunohistochemistry was used to detect neuritin expression in both NSCLC and para-cancerous tissues. The neuritin expression level was divided into low expression and high expression according to the two-grade scoring method. The relationships between neuritin expression level and clinical pathological parameters (pathological type, differentiation, tumor size, lymph node metastasis and EGFR gene) were analyzed. Results The positive particles of neuritin were located in the cytoplasm. The positive rate of neuritin in the NSCLC tissues (75.86%, 66/87) was significantly higher than that in the para-cancerous tissues (31.03%, 27/87; P〈 0.05). Neuritin expression in the NSCLC tissues was correlated with pathological type, differentiation, tumor size and EGFR gene mutation (P〈 0.05); and had no relationship with gender, age, smoking history, tumor location or lymph node metastasis (P〉 0.05). The median progression free survival (PFS) had no statistical significance between the high-expression (16 months) and low-expression (13 months) groups (P〉0.05). The median overall survival (OS) was 37 months in the neuritin high-expression group and 47 months in the neuritin low-expression group without significant difference (P〉 0.05). Conclusions Neuritin protein is expressed in both NSCLC and para-carcinoma tissues, but highly expressed in NSCLC tissues. Neuritin expression is correlated with patholog- ical type, differentiation, tumor size and EGFR gene mutation in NSCLC. It may play an important role in the occurrence and development of NSCLC.
