宫颈癌前病变组织中高危型HPV感染与原癌基因、抑癌基因表达的相关性分析

Relationship between the infection of high-risk HPV in cervical precancerous lesion and the expression of oncogene,anti-oncogene

目的:研究宫颈癌前病变组织中高危型HPV感染与原癌基因、抑癌基因表达的相关性。方法:选择2014年5月~2016年5月本院收治的宫颈癌前病变患者218例,经宫颈病理组织HPV检测分为高危型HPV组107例、低危型HPV组111例;另取同期在本院接受宫颈组织活检且证实为良性病变的100例患者作为对照组。采用RT-PCR法检测3组研究对象宫颈组织中原癌基因、抑癌基因的mRNA表达量,采用western-blot法检测宫颈组织中Sox-2及Wnt/β-catenin信号通路的蛋白表达量。结果:高危型HPV组患者的宫颈组织原癌基因DEK、Bmi-1、c-fos、K-ras、Prdx4 mRNA表达量高于低危型HPV组、对照组,抑癌基因P27、P16、DAPK、PTEN、eIF4E3 mRNA表达量低于低危型HPV组、对照组,Sox-2及Wnt/β-catenin信号通路蛋白Sox-2、β-catenin、wnt-1、wnt-3a表达量高于低危型HPV组、对照组(P<0.05)。结论:高高危型HPV感染能够通过控Sox-2及Wnt信号通路来增加宫颈癌前病变组织的原癌基因表达、减少抑癌基因表达。

Objective： To study the relationship between the infection of high-risk HPV in cervical precancerous lesion and the expression of oncogene and anti-oncogene. Methods： A total of 218 cases of cervical intraepithelial neoplasia patients in our hospital during May 2014-May 2016 were chosen and divided into high-risk HPV group（ n = 107）,low-risk HPV group（ n = 111） according to cervical tissue HPV test; Another 100 cases of patients received cervical biopsy and confirmed as benign lesions were enrolled in the control group. RT-PCR method was used to detect the mRNA expression of Proto-oncogene and anti-oncogene in three groups,Western-blot method was used to detect the protein expression of Sox-2 and Wnt / β-catenin signal pathway. Results： mRNA expression of oncogene DEK,Bmi-1,c-fos,K-ras,Prdx4 in high-risk HPV group were higher than low-risk HPV group,control group; mRNA expression of anti-oncogene P27,P16,DAPK,PTEN,eIF4E3 in high-risk HPV group were lower than low-risk HPV group,control group; expression of Sox-2 and Wnt / β-catenin signaling pathway protein Sox-2,β-catenin,wnt-1,wnt-3a in High-risk HPV group were higher than low-risk HPV group,control group（ P〈0. 05）. Conclusions： High-risk HPV infection can increase the expression of oncogene,reduce the expression of anti-oncogene in cervical intraepithelial neoplasia tissue by the regulation of Sox-2 and Wnt signaling pathway.
