摘要
目的观察人宫颈癌基因(HCCR-1)在结直肠癌、腺瘤和正常黏膜中的表达,探讨其临床意义。方法采用免疫组织化学及实时荧光定量聚合酶链反应(FQ-PCR)方法检测HCCR-1在80例结直肠腺瘤、30例结直肠癌以及20例正常结直肠黏膜组织中的表达。结果HCCR-1在正常结直肠黏膜组、结直肠腺瘤组及结直肠癌组织中表达水平的差异有统计学意义(P=0.001),阳性表达率分别为10.00%、62.50%、100.00%。在管状腺瘤、混合性腺瘤、绒毛状腺瘤组织中阳性表达率分别为46.70%、66.70%、80.00%,差异有统计学意义(P=0.015)。FQ-PCR检测结果显示在正常对照组、管状腺瘤组、混合性腺瘤组、绒毛状腺瘤组及大肠癌组HCCR-1表达量分别为0.609 2±0.326 1、4.034 6±1.188 4、12.353 4±2.828 0、22.563 7±78.952 0、43.202 9±7.675 4。结论HCCR-1在结直肠腺瘤的恶变及结直肠癌的发生发生发展过程中起一定作用。
Objective To investigate the expression and significance of human cervical canceroncogene (HCCR-1) protein in colon carcinoma, colon adenoma and normal colon mucosa.Methods Immunohistochemistry and real-time fluorescent quantitative polymerase chain reaction (FQ-PCR) were used to detect the location and expression of HCCR-1 in the 80 colon adenoma samples, 30 colon carcinoma, 20 normal colon mucosa.Results The positive expression rate of HCCR-1 protein in normal colorectal mucosa, colorectal adenoma and colorectal carcinoma tissues were 10.00%, 62.50%, 100.00%, respectively, with significant differences among them(P=0.001). The positive expression rate of HCCR-1 protein in tubular adenoma, mixed pattern of adenoma and villous adenoma were 46.70%, 66.70%, 80.00%, respectively, with significant differences among them(P=0.015). FQ-PCR showed that the HCCR-1 mRNA expression in normal colorectal mucosa, tubular adenoma, mixed pattern of adenoma, villous adenoma and colorectal carcinoma tissues were 0.609 2±0.326 1, 4.034 6±1.188 4, 12.353 4±2.828 0, 22.563 7±78.952 0, 43.202 9±7.675 4.Conclusion HCCR-1 is correlated with canceration of colorectal adenoma and the development of colorectal carcinoma.
