摘要 [目的]探究长枝木霉GF-10的固体发酵条件,并为该菌在田间的应用奠定基础。[方法]通过单因素筛选,Plackett-Burman实验和响应面分析优化该菌的产孢条件,获得影响该菌产孢的主要因素。[结果]以麸皮为基质,加入Mg SO_4 1.6‰、葡萄糖10%、(NH_4)2SO_4 1‰、Ca CO_3 1.6%、Ca SO_4 1%混合为固体培养基,光照12.17 h/d,培养47.36 h后进行搅拌,将获得最大产孢量5.93×109个/g,比优化前提高了234%;响应面分析结果显示光照和搅拌对产孢量影响最显著。[结论]该研究的完成有利于该菌的田间应用和菌剂的商业化生产。 [Aims] The aims were to explore solid fermentation conditions for the strain of Trichoderma longibrachiatum GF-10, which provide evidence for the application of this fungus in the field. [MethocIs] The solid-state fermentation conditions for the strain were studied by single factor design, Plackett-Burman design and response surface method. [Results] The results showed that the best fermentation conditions were as follows: wheat bran was used as matrix, MgSO, 1.6 ‰, glucose 10%, (NH4)2SO4 1‰, CaCO3 1.6%, CaSO4 1%, cultured with illumination for 12.17 h and stirred for 47.36 h. Under above conditions, the amount of conidia reached 5.93 × 109 cfu/g, which increased by 234% compared with single factor condition. The illumination and stir have significant effects on production of GF-10 conidia. [Conclusions] The paper laid a solid foundation for field application and commercial production.
