盐酸氨基葡萄糖对兔膝骨关节炎模型关节液中白细胞介素1和白细胞介素6及肿瘤坏死因子α水平的影响 被引量:6

摘要 目的 探究盐酸氨基葡萄糖对膝骨关节炎兔关节液中白细胞介素1(IL-1)、IL-6及肿瘤坏死因子α(TNF-α)水平的影响。 方法 选择成年雄性兔20只,随机分为 观察组和对照组,各10只。2组均制备右膝骨关节炎模型。对照组常规饮食,观察组于造模成功后在常规饮食的基础上喂食盐酸氨基葡萄糖80 mg/kg。给药喂养2个月后对比分析2组 双侧膝关节X线影像及右膝关节液中IL-1、IL-6、TNF-α水平。 结果 给药2个月后X线片显示观察组右膝关节肿胀不明显,而对照组关节肿胀明显。2组给药前右膝关节液中IL -1、IL-6、TNF-α水平比较差异无统计学意义(均P>0.05);给药2个月后,观察组IL-1、IL-6、TNF-α较给药前明显下降且明显低于同期对照组[(1.7±0.8)mg/L比(2.8± 0.8)、(3.7±0.8)mg/L,(0.82±0.27)mg/L比(1.32±0.43)、(1.98±0.46)mg/L,(2.06±0.24)mg/L比(3.08±1.17)、(4.13±1.25)mg/L](均P<0.05)。 结 论 盐酸氨基葡萄糖可降低膝骨关节炎模型兔关节液中IL-1、IL-6及TNF-α水平,进而缓解骨性病变与关节损伤。 Objective To explore the effect of glucosamine hydrochloride on interleukin 1(IL-1), IL-6 and tumor necrosis factor α(TNF-α) in synovial fluid of knee osteoarthritis in rabbits. Methods Twenty adult male rabbits were set up as right knee osteoarthritis models and divided into observation group(n=10) and control group(n=10). The control group was regularly fed; the observation group was added glucosamine hydrochloride 80 mg/kg for 2 months. Bilateral knee joints were scanned by X-ray; IL-6, IL-1 and TNF-α levels in synovial fluid of right knee joint were detected. Results Rats in observation group showed less visible swelling and inflammatory lesion of right knee joint in X-ray images compared to those in control group. Synovial fluid IL-6, IL-1 and TNF-α levels had no significant differences between groups before drug treatment(P〉0.05); the levels in observation group after drug treatment were significantly lower than those before drug treatment and those in control group[(1.7±0.8)mg/L vs (2.8±0.8),(3.7±0.8)mg/L; (0.82± 0.27)mg/L vs (1.32±0.43),(1.98±0.46)mg/L; (2.06±0.24)mg/L vs (3.08±1.17),(4.13±1.25)mg/L](P〈0.05). Conclusion Glucosamine hydrochloride can reduce IL-6, IL-1 and TNF-α levels in synovial fluid in knee osteoarthritis rabit, thus to alleviate bone lesion and joint injury.
