人尿激肽原酶对脑缺血再灌注大鼠表皮生长因子表达的影响

摘要 目的:观察大鼠脑缺血再灌注损伤后脑组织中表皮生长因子(EGF)的表达及人尿激肽原酶对其表达的影响。方法:将24只大鼠随机分为对照组(sham组)、模型组(model组)和实验组(test组),每组8只。制作大脑中动脉脑缺血再灌注模型,实验组于再灌注后5min静脉注射人尿激肽原酶3.5×10-3 PNAU/kg,在缺血2h再灌注48h后采用半定量PCR法、免疫组织化学法和Western blot法检测脑组织EGF的表达。结果:与模型组相比,实验组脑组织EGF的表达明显增高,差异有统计学意义(P<0.05)。结论:人尿激肽原酶可通过促进EGF的表达从而起到神经保护作用。 Objective:To explore the effects of human urinary kallidinogenase on the expression of epidermal growth factor(EGF)on cerebral ischemia-reperfusion injury in rats brain tissue.Methods:Rats were randomly assigned into three groups:sham operation group(sham group),focal cerebral ischemia-reperfusion group(model group)and human urinary kallidinogenase group(test group).The middle cerebral artery occlusion reperfusion model was made by the suture method(ischemia for 2hours,and reperfusion for 48hours).Rats were decapitated to detect the EGF expression in the infarct area by RT-PCR,immunohistochemistry and Western blot.Results:Compared with model group,the expression of EGF in brain tissues was increased of test group(P〈0.05).Conclusion:Human urinary kallidinogenase can protect the brain injured partly by improving the expression of EGF in brain.
