摘要
目的:建立柴胡中角鲨烯含量测定的高效液相分析方法。方法:采用ODS-C18色谱柱(250 mm×4.6 mm,5μm),流动相为甲醇∶乙腈-40∶60,检测波长为203 nm。结果:柴胡药材中角鲨烯含量为0.104 mg/g,在进样量0.2064~6.1920μg范围内呈良好线性关系(r=0.9994),平均回收率为100.78%,RSD为1.13%(n=6),重复性RSD=1.60%,精密度RSD=0.27%。结论:该方法准确可靠,可用于角鲨烯含量测定。
Objective:To establish a method for the determination of squalene in Bupleuri Radixby reverse-phase high performance liquid chromatography(RP-HPLC). Methods:The separation was performed on ODS-C18 column(250 mm×4.6 mm,5 μm)with methanol-acetonitrile(40∶60)as mobile phase and UV detector at 203 nm. Results:The content of squalene was 0.104 mg/g.The linear relationship was good for squalene in scope of 0.2064-6.1920 μg(r=0.9994). The average recovery was 100.78% with RSD=1.13%(n=6),repetition RSD =1.6% and precision RSD=0.27%. Conclusions:The method is accurate and reliable,which can be used for determining content of squalene.
