基于TGF-β1/Smad3信号通路探讨血竭含药血清对皮肤成纤维细胞的影响 被引量:9

Effects of Serum Containing Sanguis Draxonis on Skin Fibroblasts Based on TGF-β1/Smad3 Signaling Pathway

摘要 目的:探讨血竭含药血清对皮肤成纤维细胞TGF-β1/Smad3信号通路的影响。方法:将大鼠随机分为血竭高、中、低剂量含药血清组和正常对照组,每组6只。用药组分别以0.2、0.1、0.05 g·kg-1·d-1的血竭灌胃,正常对照组给予生理盐水,7 d后取血制备含药血清。组织块法培养大鼠成纤维细胞,检测血竭含药血清对成纤维细胞增殖活性、TGF-β1分泌水平以及TGF-βRⅠ、TGF-βRⅡ、Smad3和p-Smad3蛋白表达的影响。结果:与正常对照组比较,血竭各剂量含药血清能显著提高成纤维细胞的增殖活性和TGF-βRⅠ、TGF-βRⅡ及p-Smad3蛋白的表达(P<0.05或P<0.01);中、高剂量含药血清组TGF-β1水平、Smad3、和Smad3蛋白磷酸化水平明显上升(P<0.05或P<0.01)。结论:血竭对创伤的修复作用可能与其调节TGF-β1/Smad3信号转导通路有关。 Objective: To explore the effects of serum containing Sanguis Draxonis (SD) on TGF -β1/Smad3 signaling pathway of skin fibroblasts. Methods. Rats were randomly divided into high, middle, low doses of serum containing SD groups and normal control group,6 in each group. Treatment groups were given SD(0.2,0.1,0.05g·kg^-1·d^-1) by gavage and normal control group was fed by normal saline. At the end of the 7th day, serum containing SD was prepared. Rat fibroblasts were cultured by tissue explant. The effects of serum containing SD on fibroblast proliferation,the levels of TGF - β1 and the expressions of TGF - βR I ,TGF -βR Ⅱ, Smad3 and p - Smad3 were determined. Result : Compared with normal control group, the fibroblast proliferation and the expressions of TGF -βR I, TGF - βR Ⅱ and p - Smad3 were significantly raised in each dose group of serum containing SD( P 〈 0. 05 or P 〈 0. 01 ). The expressions of TGF -β1 and Smad3 and the levels of Smad3 protein phosphorylation were obviously increased in serum containing SD high and middle dose groups( P 〈 0. 05 or P 〈 0. 01 ). Conclusion : The effects of SD on wound healing may be associated with reg-ulating TGF -β1/Smad3 signaling pathway.
