摘要
目的:观察赤芍提取物对早期糖尿病肾病大鼠血糖、血脂及肾功能的影响。方法:采用单侧肾切除术和链脲佐菌素诱导的方法建立大鼠糖尿病肾病模型,并随机将模型成功的大鼠分为模型组、赤芍提取物低、中、高剂量组(剂量分别为5、10、15 g·kg-1),另取10只健康大鼠作为正常对照组,正常对照组和模型组灌胃给予蒸馏水,所有组均连续灌胃8周。观察各组大鼠8周的血糖(GLU)、血总胆固醇(TC)、甘油三酯(TG)、血尿素氮(BUN)、肌酐(SCr)动态变化。结果:1与正常对照组比,模型组8周期间的GLU均显著地升高(P<0.01),赤芍给药组8周期间的GLU均显著地升高(P<0.01),而与模型组比,均无显著性差异(P>0.05);2与正常对照组比,模型组8周期间的TC、TG均显著地升高(P<0.01),给药后2周、4周,赤芍给药组的TC、TG均较高(P<0.01)。而给药后4周时,与模型组对比,中剂量组的TC明显降低(P<0.05),高剂量组的TC也明显降低(P<0.01),3组的TG均明显降低(P<0.01)。给药后8周时,与正常对照组比,低剂量组和中剂量组的TC、TG均较高(P<0.01),高剂量组的TC、TG较高(P<0.05),而与模型组比,低剂量组的TC明显降低(P<0.05),中剂量组和高剂量组的TC也明显降低(P<0.01),3组的TG均明显降低(P<0.01);3与正常对照组比,模型组8周期间的SCr、BUN均显著地升高(P<0.01)。给药后2、4周,与正常对照组比,3组赤芍提取物给药组的SCr、BUN均较高(P<0.01)。而给药后4周时,与模型组比,低剂量组的SCr、BUN明显降低(P<0.05),中剂量组和高剂量组的SCr、BUN也明显降低(P<0.01)。给药后8周时,与正常对照组比,低剂量组和中剂量组的SCr、BUN均较高(P<0.01),高剂量组的SCr、BUN较高(P<0.05),而与模型组比,3组赤芍提取物给药组的SCr、BUN也明显降低(P<0.01)。4与正常对照组比较,模型组的肾重与体重比值均显著地升高(P<0.01)。与模型组对比,3组赤芍提取物给药组的肾重与体重比值均显著地降低(P<0.01)。5与正常对照组比较,模型组大鼠肾脏形态学呈现出以下显著改变:肾小球体积增大,肾小球纤维化,系膜区、基质增生,足细胞足突融合;3组赤芍提取物给药组大鼠肾脏病理学形态较模型组显著性改善。结论:赤芍提取物能有效地调节早期糖尿病肾病大鼠的血脂,对糖尿病肾病大鼠肾损伤有一定的保护作用。
Objective :To explore the influence of blood sugar, blood lipid and renal function in diabetic nephropathy rats by radix paeoniae rubra extract. Methods : Diabetic nephropathy model was established through the method of unilat- eral nephrectomy combined with abduction by streptozotocin. The successful diabetic nephropathy models were randomly divided into model group,radix paeoniae rubra extract low, medium and high dose groups. The radix paeoniae rubra extract low,medium and high dose groups were given 5 g·kg^-1,10 g·kg^-1 and 15 g·kg^-1 of radix paeoniae rubra extract once a day, respectively, Another healthy rats were taken as the normal control group. The normal control group and model group' s mice were given distilled water. All groups were filled the stomach for 8 weeks. Blood glucose (GLU) ,total cholesterol (TC),triglyceride (TG), blood urea nitrogen (BUN) and creatinine (SCr) in rats were determined. Results: ( l ) Compared with normal control group, GLU of model group was significantly increased during eight weeks ( P 〈 0. 01 ). GLU in radix paeoniae groups was significantly higher during eight weeks ( P 〈 0. 01 ) and compared with model group, there was no significant difference ( P 〉 0.05 ). ( 2 ) Compared with normal control group, TC and TG of model group were significantly increased during eight weeks (P 〈 0. 01 ). 2 and 4 weeks after treatment, TC and TG of radix paeoniae groups were higher (P 〈 0. 01 ). And at 4 weeks after treatment, compared with model group, TC of medium dose group was significantly decreased ( P 〈 0. 05 ) and that of high dose group obviously lower( P 〈 0. 01 ). TG in three doses groups were significantly lower ( P 〈 0. 01 ). 8 weeks after treatment, compared with normal control group,TC and TG of low and middle dose groups were higher ( P 〈 O. 01 ). Those of high dose group were higher( P 〈 0. 05 ) and compared with model group, TC of low dose group was significantly decreased ( P 〈 0. 05 ) and that of medium and high dose groups were also significantly reduced (P 〈 0. O1 ). TG in three doses groups were significantly lower (P 〈 0. 01 ). (3)Compared with normal control group, SCr and BUN of model group were significantly increased during eight weeks (P 〈 0. 01 ). 2 and 4 weeks after treatment, compared with normal control group ,SCr and BUN of three doses groups were higher (P 〈 0. 01 ). And 4 weeks after delivery, compared with the model group, SCr and BUN of low dose group were significantly decreased ( P 〈 0. 05) and those of medium and high dose groups were also significantly reduced ( P 〈 0. 01 ). At 8 weeks after treatment ,compared with normal control group, SCr and BUN of low and medium doses groups were higher (P 〈 0. 01 ) and those of high dose group were higher (P 〈 0.05) and compared with model group, SCr and BUN of three doses groups were also significantly reduced ( P 〈0. 01 ). (4) Compared with normal control group ,the ratio of kidney weight/weight in model group was significantly higher (P 〈 0. 01 ). Compared with model group, the ratio of three doses groups were sig- nificantly higher (P 〈 0. 01 ). (5) Compared with normal control group, model group' s rats followed significant kidney morphology changes : glomerular volume increased, glomerular fibrosis, mesangial area and matrix hyperplasia, podocyte foot process fusion. Compared with model group, those in three doses groups were significantly improved. Conclusion:Ra- dix paeoniae rubra extract can effectively adjust the blood lipid of early diabetic nephropathy rats and has certain protective effect for renal damage of diabetic nephropathy rats.
