摘要
目的建立同时测定银翘马勃散中绿原酸、连翘酯苷A、牛蒡苷、连翘苷、次野鸢尾黄素5种成分含量的方法。方法采用HPLC法。COSMOSIL 5 C_(18)-AR-Ⅱ色谱柱(4.6 mm×250 mm,5μm);以甲醇-0.4%磷酸水为流动相,梯度洗脱;流速:1.0 m L·min^(-1);柱温:30℃;检测波长:272 nm。结果绿原酸、连翘酯苷A、牛蒡苷、连翘苷、次野鸢尾黄素的保留时间分别为24.963,39.996,45.863,47.983,63.868 min;线性范围分别为0.992~9.92(r=0.9999),0.498~4.98(r=0.9996),2.66~26.6(r=0.9999),0.092~0.92(r=0.9998),0.0149~0.149μg(r=0.9997);平均加样回收率分别为99.82%、96.38%、96.69%、103.23%、98.70%。结论该方法简单、准确、稳定,可用于银翘马勃散中绿原酸、连翘酯苷A、牛蒡苷、连翘苷、次野鸢尾黄素的同时测定,为银翘马勃散及其类方制剂的新药开发与质量评价提供参考。
Objective To establish a HPLC method for simultaneous determination of chlorogenic acid,forsythiaside A,arctiin,phillyrin and irisflorentin in Yinqiao Mabo Powder. Methods HPLC was performed on a COSMOSIL5C(18)-AR-Ⅱ column(4.6 mm×250 mm,5 μm) with methanol-0.4 % phosphoric acid as mobile phase in gradient elution,the flow rate was 1.0 m Lomin-1,the column temperature was set at 30 ℃,and the detection wave length was 272 nm. Results The linear range of chlorogenic acid,forsythiaside A,arctiin,phillyrin and irisflorentin was0.992-9.92(r=0.9999),0.498-4.98(r=0.9996),2.66-26.6(r=0.9999),0.092-0.92(r=0.9998),0.0149-0.149 μg(r=0.9997) respectively, and their average recovery was 99.82 %, 96.38 %, 96.69 %, 103.23 %, 98.70 %respectively. Conclusion The method is convenient,accurate and stable for simultaneous determination of chlorogenic acid,forsythiaside A,arctiin,phillyrin and irisflorentin in Yinqiao Mabo Powder,which will supply evidence for the development and quality control of Yinqiao Mabo Powder and its relevant preparations.
