山大颜种子无菌萌发实验研究 被引量:1

Experiment Study on Aseptic Germination of Psychotria rubra(Lour.) Poir. Seeds

摘要 目的建立基于组织培养技术的山大颜种子无菌萌发方法。方法观察、采集、记录山大颜果实、种子特征;以不同采收时期新鲜剥去种皮的种子为实验材料,分别进行无菌萌发实验,考察山大颜种子最佳采收时期;以剥去种皮的山大颜成熟种子为外植体,筛选适宜的外植体消毒时间;考察山大颜种子在4种不同培养基上的萌发情况;采用L9(34)正交试验设计法考察6-苄氨基腺嘌呤(6-BA)、赤霉素(GA)、α-萘乙酸(NAA)3种激素对种子萌发的影响。结果 9月份采收的山大颜种子活性较强,可以用作实验材料;2.0 g·L^(-1)升汞溶液浸泡时间2 min的种子生长率高、污染率较小,适合无菌培养;山大颜种子萌发的最优培养基为MS+2.0 mg·L^(-1)6-BA+0.1 mg·L^(-1)GA+3.0 g·L^(-1)活性炭。结论该方法建立了山大颜种子无菌萌发的适宜条件,有助于提高山大颜的繁殖率,为山大颜种质的人工扩大繁殖提供参考。 Objective To establish a method for the aseptic germination of Psychotria rubra(Lour.)Poir. seeds based on the plant tissue-culture technique. Methods We collected the fruits and seeds of Psychotria rubra(Lour.)Poir. for the study of their characteristics. Fresh peeled seeds harvested at different time were used as the experimental material for aseptic germination experiment to optimize the harvesting time. The germination of the seeds planted in four kinds of culture medium was investigated. And L9(34)orthogonal experimental design was adopted for evaluating the effects of three hormones of 6- 6-benzylaminopurine(6-BA),gibberellin(GA),α-naphthylacetic acid(NAA)effects on seed germination. Results The results showed that Psychotria rubra(Lour.) Poir. seeds harvested in September had the strongest activity,and can be used as the experimental material. The seeds soaked in 2.0 g·L-1mercuric chloride solution for 2 min were suitable for cultivating and planting,which resulted into high growth rate and low pollution rate.The optimal culture medium for the germination of Psychotria rubra(Lour.) Poir. seeds was MS +2.0 mg·L-16-BA +0.1 mg·L-1GA +3.0 g·L-1activated charcoal. Conclusion The optimal conditions for the aseptic germination of Psychotria rubra(Lour.) Poir. seeds have been established, which are helpful to improve the reproduction rate of Psychotria rubra(Lour.) Poir.,and will provide reference for artificial propagation of Psychotria rubra(Lour.)Poir..
