摘要 目的研究神经鞘氨醇磷酸胆碱(shingosylphosphorylcholine,SPC)对体外培养人真皮成纤维细胞分泌基质金属蛋白酶-1(MMP-1)、基质金属蛋白酶-3(MMP-3)、基质金属蛋白酶内源性组织抑制剂-1(TIMP-1)的影响,探索在促进皮肤创伤愈合方面SPC的作用机制。方法通过MTT的方法检测SPC在成纤维细胞增殖效率方面的作用。利用RT-PCR和Western blotting的方法检测体外培养人真皮成纤维细胞经5μmol/L SPC诱导后,表达损伤修复相关蛋白MMP-1,MMP-3,TIMP-1的情况。结果一定浓度的SPC具有促进成纤维细胞增殖的作用。5μmol/L SPC具有上调成纤维细胞表达MMP-1,MMP-3,TIMP-1 m RNA和蛋白的作用(P<0.05)。结论实验证实体外培养人真皮成纤维细胞在经SPC干预后,其表达细胞外基质调节蛋白MMP-1,MMP-3,TIMP-1的作用得到增强,提示SPC具有促进皮肤创伤愈合的作用。 Objective To investigate the effect of sphingosylphosphorylcholine (SPC) on expression of MMP-1, MMP- 3, TIMP-1 in human dermal fibroblasts in vitro, and elucidate the role of SPC in skin wound healing. Methods The proliferation of fibroblasts was assessed by MTT assay. The expressions of MMP-1, MMP-3, TIMP-1 in human dermal fibroblasts, which were stimulated with 5 μmol/L SPC, were detected by using RT-PCR and Western blotting. Results SPC could promote the fibroblast proliferation. Meanwhile, SPC induced the expression of MMP-1, MMP-3, TIMP-1 in mRNA and protein level (P 〈 0. 05 ). Conclusion Sphingosylphosphorylcholine (SPC) could enhance the expression of extracellular matrix regulatory proteins, such as MMP-1, MMP-3 and TIMP-1 in human dermal fibroblasts in vitro, suggesting that SPC might play a role in promoting skin wound healing.
