摘要 肿瘤由基因、环境、代谢及信号通路异常等多种致病因素共同作用引起,这些异常可严重扰乱细胞增殖与死亡间的稳态平衡。一般而言,高选择性药物可特异性阻断一种信号通路,但发挥治疗作用有限,因此,通过多位点、多通路有效抑制肿瘤细胞的药物成为研究新方向。甘草查尔酮A(Licochalcone A,LCA),一种新型黄酮类化合物,是甘草根部的主要活性提取物,具有多种生物学活性,例如抗氧化、抗病毒、抗炎、抗菌、抗疟疾、抗血管生成及骨诱导性,其抗肿瘤特性受到广泛关注,可通过促凋亡、阻滞细胞周期及抗侵袭与转移等多种作用参与杀伤肿瘤细胞。本文拟阐述LCA的抗肿瘤效应及其分子机制,为进一步临床研究提供参考。 Cancer is a multi- pathogenic disease comprised of genetic,epigenetic,metabolic and signaling aberrations which severely disrupt the normal homoeostasis of cell growth and death. Rational developments of highly selective drugs which specifically block only one of the signaling pathways have been associated with limited therapeutic success. Multi- targeted suppression of cancer has emerged as a new direction for effective anti- cancer treatment.Licochalcone A,a newly chalconoid,is one of the major active components of the roots of Glycyrrhiza species and possesses multiple biological and pharmacological properties including,antioxidant,antiviral,anti- inflammatory,antimicrobial,antimalarial,antiangiogenic,and osteogenic activities. Recently,the anti- cancer activity of Licochalcone A has been extensively studied,it has been shown to fight cancer by inducing apoptosis,cell cycle arrest,and inhibiting invasion and metastasis by targeting multiple signaling pathways. The purpose of this review was to discuss its anti-tumor activity and molecular mechanisms which are aimed at the further designing and conducting of clinical trials.
