摘要
建立了用高效液相色谱法快速检测果酒中有机酸的方法。最佳色谱条件:色谱柱为Thermo C_(18)反相柱(4.6×250mm,5μm),pH为2.55、0.01mol/L的NH_4H_2PO_4缓冲溶液为流动相,洗脱流速1.2mL/min,色谱柱温45℃,检测波长为210nm;洗脱时间9min。在此条件下,10种有机酸得到了良好的分离。该方法具有较好的线性关系(R〉0.998 24)和较宽的线性范围,相对标准偏差(RSD)为0.25%~3.84%,加标回收率为77.27%~104.56%,重现性好,精确度高。将此方法应用于青梅酒、樱桃酒、蓝莓酒、枇杷酒、荔枝酒、葡萄酒和桑葚酒中有机酸的测定,得到的结果准确可靠,可以用于果酒品质的控制和工艺的调整。
A rapid method for determination of organic acids in fruit wines is established.The optimal conditions are as follows:Thermo C(18) reversed-phase column(4.6×250mm,5μm)is used;0.01mol/L NH4H2PO4 of pH 2.55 is used as mobile phase;flow rate is 1.2mL/min;the column temperature is set at 45℃;and measurement wavelength is 210 nm.10organic acids are separated well in 9 minutes.The peak areas and concentrations show good correlations(R0.998 24).The method has good reproducibility and high accuracy when RSD is in the range of 0.25%-3.84%(n=5)and the recovery is 77.27%-10~4.56%.Organic acids in plum wine,cherry rum,blueberry wine,loquat wine,lychee wine,red wine and mulberry wine can be determined by this method,and the satisfactory results can be obtained.
