摘要
探讨mir-187对胃癌细胞恶性生物学行为的影响;采用q PCR检测miR-187在胃癌SGC7901细胞、MKN45细胞和人永生化胃黏膜上皮细胞GES细胞中的表达,应用miR-187 mimics转染SGC7901细胞和MKN45细胞,通过MTT实验、流式细胞术和裸鼠成瘤实验检测miR-187对胃癌细胞增殖、凋亡的影响。结果 SGC7901、MKN45细胞中miR-187的表达明显高于GES细胞;与对照细胞相比,MTT实验显示转染miR-187 mimics后胃癌SGC7901细胞的增殖增快(P<0.05),流式细胞检测表明miR-187mimics转染可以减少SGC7901细胞的凋亡(P<0.05),裸鼠成瘤实验提示转染miR-187mimics可以促进SGC7901细胞成瘤(P<0.05)。说明miR-187可以促进胃癌细胞生长、抑制凋亡和促进增殖,提示miR-187在胃癌恶性生物学行为中发挥重要作用,有可能成为诊断胃癌的新标志和治疗胃癌的新靶点。
To investigate the effect of miR-187 on the malignant behaviours of gastric cancer cells,q PCR was performed to detect the expression of miR-187 in gastric cancer cells SGC7901,MKN45 and the human immortal gastric epi-thelial cell GES cells. miR-187 mimics was transfected into SGC7901 and MKN45 cell. MTT assays,flow cytometry assay,tumorigenicity in nude mices were performed to evaluate the effect of miR-187 on the cell growth,cell apoptosis and proliferation of gastric cancer SGC7901 cells. The expression of miR-187 in SGC7901 and MKN45 cells were higher than that in GES cells. As indicated by MTT assay,compared with control cells,miR-187 mimics transfected SGC7901 and MKN45 cells showed a significantly increased rate of cell proliferation.Flow cytometry assay showed a decreased apoptotic index of miR-187 mimics transfected SGC7901 and MKN45 cells. Tumorigenicity in nude mices suggested miR-187 could promote the proliferation of SGC7901 and MKN45 cells in vivo. It is conclusied that MiR-187 promotes the growth,proliferation of SGC7901 and MKN45 cells,and it can also regress the apoptosis of gastric cells. These results suggest miR-187 plays an important role in the malignant phenotypes of gastric cancer. MiR-187 could be used to the new marker in the diagnosis of gastric cancer and the new target in the treatment of gastric cancer.
