rhEPO对尿毒症患者氧化应激水平及内皮细胞凋亡的影响 被引量:1

Effects of rhEPO on oxidative stress levels and endothelial cell apoptosis of patients with uremic serum

摘要 目的:探索rhEPO对尿毒症患者氧化应激水平及内皮细胞凋亡的影响。方法选取2015年1月至2016年1月新乡医学院第三附属医院收治的尿毒症患者9例,以及社会健康志愿者9例,按内皮细胞不同的作用方式分为对照组(含10%健康者血清)、尿毒症组(含10%尿毒症患者血清)、rhEPO组(培养基含有5、10、15 U/ml三种不同浓度的rhEPO和10%尿毒症患者血清)三组。各组培养作用于内皮细胞24 h后,采用比色法测定细胞内活性氧、DNA断裂的原位末端标记法检测细胞凋亡。结果 rhEPO可降低细胞内氧化应激水平,抑制尿毒症患者血清诱导的内皮细胞凋亡(P〈0.05),此外,当rhEPO浓度增大时,氧化应激水平及内皮细胞凋亡指数均逐渐降低(P〈0.05);方差相关性分析结果表明氧化应激水平及内皮细胞凋亡指数呈很明显的正相关性关系(r=0.89,P〈0.01)。结论 rhEPO在一定程度上可以抑制尿毒症患者血清诱导的内皮细胞凋亡,该结果是通过减轻氧化应激反应而产生的。 ObjectiveEffects of rhEPO on oxidative stress levels and endothelial cell apoptosis of patients with uremic serum.MethodsFrom January 2015 to January 2016, 9 cases patients with uremic serum from the Third Affiliated of Xinxiang Medical Hospital and 9 cases of healthy volunteers were selected in this study. According to the effect of different methods to HUVECs, HUVECs were divided into 3 groups: control group (containing 10% health serum medium), uremia group (containing 10% uremic patients serum medium), rhEPO group (medium containing 5, 10, 15 U/ml three different concentrations of rhEPO and 10% uremic patients serum medium medium) and 10% of patients with uremia. After Each medium of group culture effecting on endothelial cells 24 h, reactive oxygen species were detected by colorimetry and cell apoptosis was evaluated by TUNEL assay.Results rhEPO could reduce oxidative stress levels in cells, and had significant inhibition against apoptosis of endothelial cells induced by uremia serum (P〈0.05), in addition, when the concentration of rhEPO increases, the oxidative stress levels and endothelial cell apoptosis index were reduced gradually (P〈0.05). The results of variance correlation analysis showed that the oxidative stress levels and endothelial cell apoptosis index were obviously positive correlation relations (r=0.89,P〈0.01). Conclusion rhEPO could significantly inhibit the uremia serum induced apoptosis of endothelial cells, the mechanism may be related to the rhEPO which can reduce the oxidative stress reaction.
