摘要
裂殖壶菌细胞结构紧密,对其进行破壁是二十二碳六烯酸(DHA)高强度发酵的重点工序。以裂殖壶菌为实验菌种,比较了水洗法、膜过滤法、高压均质法、酶解法和膜过滤高压均质法5种破壁方法,观察了每种方法破壁前后细胞形态特征,同时测定细胞破壁率、发酵液含油量、DHA产量和DHA含量。结果表明:膜过滤高压均质法效果最为理想,破壁率达到100%;膜过滤高压均质法的含油量、DHA产量、DHA含量均为最高,分别达到16.98 g/L、7.08 g/L、41.71%;从生产的连续性和有效性来讲,膜过滤高压均质法是最佳的破壁方法。
Schizochytrium' s cell structure is tight, so wall - breaking is the key process of DHA high strength fermentation. With Schizochytrium as experimental bacteria, the wall - breaking methods, including water - washing , membrane filtration, high pressure homogeneous, enzymolysis and high pressure homogeneous composite membrane filtration were compared, and the morphological characteristics of cells before and after wall - breaking by each method were observed, in the same time the cell wall - breaking rate, oil content of fermentation liquid, and productivity and content of DHA were determined. The results showed that the effect of high pressure homogeneous composite membrane filtration method was the best with wall - breaking rate 100%, also the oil content of fermentation liquid, and productivity and content of DHA of that method were the highest, reaching 16.98,7.08 g/L and 41.71% ,respectively. So high pressure homogeneous composite membrane filtration method was the best way to break the wall from the aspect of continuity and efficiency of production.
